大鼠大脑中动脉栓塞模型的建立

摘    要:目的:建立大鼠大脑中动脉栓塞(MCAO)模型,以便今后进一步进行缺血性脑梗死方面的研究。方法:将大鼠随机分成模型组和假手术组,采用线栓法建立大鼠左侧MCAO模型,术后观察体征,进行神经功能缺损评分、脑组织病理学检查、磁共振成像(MRI)检查、血清抗氧化指标测定。结果:假手术组大鼠全部存活,模型组除2只因麻醉过度死亡、1只插线未成功外,其余7只均出现左侧Horner征,神经功能缺损评分在1~3分之间。模型组大鼠脑组织可见显著病理改变,MRI检查脑组织缺血病变明显。模型组血清SOD和GSH-Px活性高于假手术组,MDA含量低于假手术组,差异均有统计学意义(P<0.05)。结论:成功建立了大脑中动脉栓塞模型。

Abstract:Objective:To establish a cerebral ischemia animal model induced by middle cerebral artery occlusion(MCAO) in rats so that to carry out the research of ischemia cerebral infarction. Methods:Sprague-Dawley rats were randomly assigned into two groups: Model and Sham operation(OP). Middle artery occlusion was applied by thread technique in all rats in Model group. The symptoms,the function of nervous system and the pathology change of brain were observed after the operation. Ischemic penumbra was observed by MRI,too. The antioxicidative indexes were measured. Results: The rats in Sham OP group was all alive. There were two rats in Model group died in anesthesia and the operation in one rat in Model group was not be completed successfully. The rest 7 rats all had the symptoms of Horner in left side,and the nervous system function score was 1 to 3. Obvious pathology changes were found in Model group. There were great changes in Model group by MRI. The activities of serum SOD and GSH-PX in Model were significantly lower than those in Sham OP. The contents of serum MDA were significantly higher than those in Sham OP. Conclusion:MCAO is success.This technique could be used in the establishment of MCAO model.
