热疗联合DDP诱导大肠癌细胞凋亡和bcl-2表达的实验研究

摘    要:目的 :研究热化疗诱导大肠癌 (LoVo)细胞凋亡及其与bcl_2基因表达的关系 ,探讨热化疗治疗大肠癌的机制。方法 :应用热疗联合顺铂 (DDP)在不同药物浓度、时间和温度条件下诱导LoVo细胞凋亡 ,以流式细胞仪检测其凋亡率及bcl_2基因的表达。结果 :随着药物浓度的升高、作用时间的延长 ,细胞凋亡率增加 ;热疗联合DDP所致凋亡率大于单纯化疗组和单纯热疗组之和 ;随着药物浓度的升高 ,bcl_2的表达下调 ;各处理组与对照组比较差异有显著性 (P <0 .0 5 ) ;热化疗使bcl_2表达下调较单纯热疗及单纯化疗显著 (P <0 .0 5 ) ;高温也可以使bcl_2的表达下调。结论 :单纯化疗、热化疗及单纯热疗均能诱导LoVo细胞凋亡且可使bcl_2基因表达下调 ;热疗联合DDP在诱导凋亡及下调bcl_2基因表达方面具有协同作用。

Abstract:Objective:To study the roles of hyperthermo-chemotherapy in human colonic cancer cell apoptosis and related gene expression.Methods:we used hyperthermia combined with DDP,with different dose,time and temprature to induce Lo Vo cell line apoptosis,then to assay apoptosis rate and bcl-2 gene expression by flow cytometry.Results:With the elevation of dose and time,the apoptosis rate induced by the drug increased.Hyperthermia combined with DDP group have a higher apoptosis rate than chemotherapy and hyperthermotherapy alone,it showed that hyperthermia combined with DDP has a synergism.Following with the elevation of dose,expression of bcl-2 descend.There is a significant difference between the test group and the contrast group (P<0.05);hyperthermia also make a low expression of bcl-2 gene.Hyperthermia combined with DDP caused a significant inhibition of expression of bcl-2 than hyperthermia and chemotherapy alone (P<0.05 respectively).Conclusion:DDP and single hyperthermia could induce LoVo cell apoptosis and decrease the expression of bcl-2 gene;hyperthermotherapy combined with DDP has a symergism in inducing apoptosis and inhibit bcl-2 expression.
