酢浆草遗传多样性的ISSR分析

摘    要:目的 探讨酢浆草种质资源遗传多样性,为酢浆草遗传差异的评定及优良种质资源的筛选提供分子水平的参考依据.方法 采用简单序列重复区间(ISSR)分子标记技术对酢浆草18个居群36个样本及红花酢浆草2个居群4个样本进行遗传多样性分析.结果 从59条ISSR引物中筛选出10条引物用于PCR扩增,共扩增出89条DNA条带,其中多态性条带78条,占87.64%,平均每个引物扩增条带的数目为8.9条.利用POPGENE1.32和NTSYS-pc软件分析得出40个样本的平均有效等位基因数为1.3888,平均Nei''s基因多样性指数为0.3493,平均Shannon信息指数为0.2252;UPGMA聚类结果,在遗传相似性系数0.6558处把供试的40个样本分为4大类.结论 本研究选用的酢浆草样本遗传多样性比较丰富,酢浆草与红花酢浆草的ISSR遗传多样性差异较明显.

ISSR analysis on genetic diversity of Oxalis
Abstract:Objective To study the genetic diversity of germplasm resources of Oxalis corniculata,and to provide molecular basis for reference of Oxalis corniculata by assessing genetic differentiation and selecting of excellent germplasm resources.Methods The genetic diversity with 36 samples of 18 populations of Oxalis corniculata and 4 samples of 2 populations of Oxalis corymbosa was analyzed by ISSR molecular marker technique.Results 10 primers were selected from 59 ISSR primers which amplified 89 DNA bands,and 78 bands among them were polymorphic which the percentage of polymorphic bands was 87.64%,the average DNA bands amplified by each primer were 8.9 bands.The average number of effective allele was 1.388 8 of 40 samples,the Nei''s gene diversity and Shannon''s information index were 0.349 3 and 0.225 2 by POPGENE1.32 and NTSYS-pc software.These 40 samples were clustered into 4 groups at the genetic similarity coefficient 0.655 8 by UPGMA.Conclusion The genetic diversity in 40 samples of Oxalis corniculata collected in this paper isrich,and there were significant differences in genetic diversity of ISSR between Oxalis corniculata and Oxalis corymbosa.
