摘    要:目的 探讨miRNA-331对大肠癌细胞HCT116增殖、迁移和侵袭等生物学功能的影响.方法 将HCT116细胞转染miRNA-331 mimics后,通过MTT实验、克隆形成实验检测miRNA-331对大肠癌细胞增殖的影响;Transwell实验检测miRNA-331对HCT116细胞迁移和侵袭的影响.结果 转染miRNA-331 mimics的HCT116细胞在MTT实验中第1天~第5天的吸光度值与对照组相比显著减低;克隆形成实验中,转染miRNA-331 mimics后,HCT116细胞形成克隆的能力被削弱;Transwell实验显示,与阴性对照相比,转染miR-NA-331 mimics后,HCT116细胞的迁移能力明显下降.结论 miRNA-331表达上调可抑制HCT116细胞的增殖、迁移能力,表明miRNA-331有潜在的抑癌作用.

The biological function of miRNA-331 in colorectal cancer HCT116 cells
Abstract:Objective To explore effects of miRNA-331 on proliferation,migration and invasion of human colorectal cancer cell line HCT116.Methods After transfection of HCT116 cells with miRNA-331 mimics,the effect of miRNA-331 on the proliferation of colorectal cancer cells was detected by MTT assay and clone formation assay.Transwell was used to detect the effects of miRNA-331 on migration and invasion of HCT116 cells.Results The absorbance values were significantly lower compared to the control group after HCT116 cells ransfected with miRNA-331 mimicsin from day 1 to day 5.After the transfection of miRNA-331 mimics,the ability of HCT116 cells to form clones was reduced.Transwell assay revealed that compared with the negative control after transfection of miRNA-331 mimics,the migration ability of HCT116 cells decreased significantly.Conclusion The up-regulation of miRNA-331 expression can inhibit the proliferation and migration of HCT116 cells,suggesting that miRNA-331 is a potent tumor suppressor.
