跨膜丝氨酸蛋白酶4在胃癌组织中的表达及其临床意义

目的 探索跨膜丝氨酸蛋白酶4(TMPRSS 4)在胃癌组织中的表达情况及与胃癌病理学特征的关系,以及明确TMPRSS 4表达与胃癌临床预后的关系.方法 使用RT-qPCR技术和免疫组化检测TMPRSS 4在115例胃癌组织样本(胃癌及癌旁组织)中的表达情况;分析TMPRSS 4表达水平与胃癌病理学特征和预后的关系.结果 RT-qPCR结果提示胃癌组织中的TMPRSS 4 mRNA水平显著高于其在匹配癌旁组织中的表达水平,差异有统计学意义(P＜0.05).免疫组化结果显示TMPRSS 4在胃癌组织中的表达比在良性组织高(56.5％vs19.5％,P＜0.001).TMPRSS 4的过度表达与胃肿瘤的分化程度、淋巴道转移及TNM分期肿瘤显著相关(P＜0.01).TMPRSS 4是影响胃癌总体生存率及无病生存率的独立因素.结论 TMPRSS 4高表达与胃癌患者相关临床病理特征及患者的预后密切相关,TMPRSS 4有可能作为一个常规的胃癌的生物标志物,在一定程度上预测肿瘤的侵袭转移.

The expressed of TMPRSS 4 in gastric cancer tissues and its clinical significance

Objective To investigate the relationship between the expression of transmembrane serine protease 4 (TMPRSS 4) and the clinicopathological features of gastric cancer (GC),and to clarify the relationship between TMPRSS 4 expression and the prognosis of GC.Methods RT-qPCR and immunohistochemical methods were used to detect the expression of TMPRSS 4 in 115 cases of GC,and the relationship between the expression level of TM-PRSS 4 in tumor tissueand the clinicopathological characteristics of GC was studied.In addition,the impact of TMPRSS 4 expression on the survival of GC was also analyzed.Results RT-qPCR results revealed that TMPRSS 4 mRNA was higher in gastric cancer tissues than those in normal gsatric tissues (P ＜ 0.05).The immunohistochemical results also indicated that the expression of TMPRSS 4 in gastric cancer tissues was higher than in benign tissue (56.5％ vs 19.5％,P ＜0.001).In addition,TMPRSS 4 was also found positively correlated with gastric cancer differentiation,lymph node metastasis and high TMN staging of tumor(P ＜ 0.01).TMPRSS 4 was an independent factor that affects the overall survival rate and disease-free survival rate of gastric cancer patients.Conclusion High expression of TMPRSS 4 is closely related to the clinicopathological features of patients with gastric cancer and the prognosis of patients.TMPRSS 4 may be a biomarker of routine gastric cancer and can predict tumor invasion and metastasis to a certain extent.
