抑癌基因nm23-H1对化疗药物增敏作用的体外实验研究

摘    要:目的:探讨抑癌基因nm23-H1对几种常用化疗药物的增敏作用。方法:构建含有人全长的nm23-H1基因cDNA的真核表达载体质粒pcDNA3-nm23-H1,通过脂质体介导,将它和空载体质粒pcDNA3分别转染鼠肺腺癌LA-795细胞株中,得到的阳性转染子分别命名为pcDNA3-nm23-H1-795细胞,pcDNA3-795细胞,另设795细胞为正常对照组,用MTT法测定nm23-H1对顺铂,氮芥,丝裂霉素C,5-氟尿嘧啶,长春新碱和足叶乙苷的增敏效应。结果:nm23-H1基因在pcDNA3-nm23-H1-795细胞中获得了表达,MTT法检测nm23-H1对顺铂,氮芥,丝裂霉素C有不同程度的增敏作用,而对5-氟尿嘧啶,长春新碱和足叶乙苷没有此效应。结论:nm23-H1可增加化疗药物顺铂,氮芥,丝裂霉素C对LA-795细胞的敏感作用。

Abstract:Objective: To investigate the sensitivity enhancing effect of tumer suppressor gene nm23 H1 to chemotherapeutic agents. Methods: Recombinant eukaryotic expression vector pcDNA3 nm23 H1 containing full length human nm23 H1 cDNA was constructed and then transfected into mice lung carcinoma cell line LA 795. Sensitivites of the cell lines to cisplatin, mechlorethamine, mitomycin C, 5-fluorouracil, vincristine and vp 16 were determined by MTT assay. Results: nm23 H1 gene was successfully transfected into 795 cells and highly expressed. MTT assay showed that nm23 H1 could enhance the sensitivity of the cell line to cisplatin, mechlorethamine and mitomycin C but not to 5-fluorouracil, vincristine and vp 16. Conclusions: nm23 H1 enhances the sensitivity of LA 795 cell line to some chemotherapeutic agents.
