摘    要:目的 探讨肺微血管内皮细胞粘附分子表达在脓毒症小鼠肺损伤中的作用。方法 盲肠结扎穿孔(CLP)制造小鼠脓毒症模型,分别在致伤后3、12h分离肺微血管内皮细胞,用逆转录-聚合酶链(RT-PCR)方法定量检测E-选择素、细胞间粘附分子(ICAM)-1和血管细胞粘附分子(VCAM)-1的基因表达,同时测定肺组织中髓过氧化物酶(MPO)活性的改变。结果 CLP后3h肺微血管内皮细胞E-选择素、ICAM-1

Abstract:Objective\ To investigate the role of adhesion molecule expression in sepsis induced lung injury in mice. Methods\ Sepsis was induced in mice by cecal ligation and puncture (CLP). Sham operation group underwent the same manipulation but without CLP. The gene expression of adhesion molecules (E selectin, ICAM 1 and VCAM 1) in lung microvascular endothelial cells were quantitatively assessed by RT PCR. The change of myeloperoxidase enzyme (MPO) activity in lung tissue was also detected. Results\ Significant increases of E selectin, ICAM 1 and VCAM 1 expression and MPO activity in lung tissue were observed at 3 h after CLP ( P < 0.01 ). At 12 h, E selectin gene expression was obviously decreased without significant different from sham operation group, while the expression of ICAM 1 and VCAM 1 genes was further increased ( P <0.01); The MPO activity in lung tissue was markedly decreased ( P <0.01) with a significant difference from sham operation group ( P <0.01). Conclusion\ The upregulation of the expression of adhesion molecules in lung microvascular endothelial cells is an important step for the migration of leukocytes to and accumulation at the site of inflammation, which might play a critical role in lung injury during sepsis.
