嘌呤霉素肾病鼠足突形态改变的形态计量学研究

摘    要:目的:定量研究嘌呤霉素肾病大鼠模型从建立到恢复过程中,足突的形态变化规律。方法:用透射电镜摄片,图像分析仪分析并应用形态计量学的方法,研究了33例大鼠(嘌呤霉素注射实验组及生理盐水注射对照组)不同时间点的足突形态变化。结果:注射嘌呤霉素后,足突表现出由变宽到融合又逐渐恢复的形态变化,形态计量学的结果进一步表明,各时间点实验组足突宽度,体积密度,比表面与对照组相比有高度显著性差异(P<0.01),表面积密度在2--15天与对照组差异显著(P<0.05)。且实验组内相邻的不同时间点间比较, 足突宽度、体积密度、表面积密度、 比表面差异均具有高度统计学意义(P<0.01)。结论:形态计量学对嘌呤霉素肾病足突形态研究,定量表现出足突由变宽到融合以后逐渐恢复的动态过程,为今后对功能与分子水平的研究提供早期准确的实验形态依据。

Abstract:Objective: To investigate the morphometric changes in the podocyte foot proce-ss during the development of, and recovery from, acute puromycin aminonucleoside (PAN) nephropathy in rats. Methodology: A total of 33 rats was studied, there were 15 rats fr-om control group and 18 from experimental group with PAN injection. Morphometric met-hods were applied, and transmission electron microscopy (TEM) as well as image analysis was used to study the changes of foot process. Result: In the process of development of PAN nephropathy, the foot process became broader and then fusion. After 10 days, the f-oot processes began to recover. The morphometric parameters indicated that the foot proc-ess width, volume density, and specific surface (ratio of surface to volume) of foot proce-ss were obviously different (P<0.01) between the experimental and control groups among different time points. The surface density was different from 2 days to 15 days between experimental and control groups (p<0.05). In experimental group, the morphometric param-eters showed significant differences among different time points (p<0.01). Conclusion: The remarkable morphological changes were revealed with morphometric method in PAN ne-phropathy rats, which would be benefit to the further functional and molecular studies.
