维吾尔族妇女子宫颈癌中 p53 基因的表达和 HPV DNA 检测研究

摘    要:应用E6特异性引物PCR检测新疆维吾尔族妇女子宫颈癌活检组织标本中的HPV6,16和18DNA。并应用免疫组化研究p53基因的表达。结果显示,65例标本中7例不适应用于PCR,其余58例中HPV16DNA阳性45例(77.6%),而HPV6DNA和18DNA为阴性。免疫组化显示,65例中p53表达阳性49例(75.4%)。在45例HPVDNA阳性标本中,p53表达阳性34例(75.6%);13例HPVDNA阴性标本中,p53表达阳性11例(84.6%)。提示,HPV阳性或阴性与p53表达之间无显著性差异(P>0.05)。

Abstract:Cancer of the uterine cervix is one of the most common malignancies in women worldwide. Strong evidence supports that human papillomavirus (HPV) infection, especially HVP types 16 and 18, may be the role in the genesis of these tumors. HPV 16 and 18 both included two transforming gene products, the E6 and E7 protein. Action of these protein may be mediated by their interaction with a tumor suppresser gene. The p53 protein plays an important role in controlling proliferation of neoplastic and normal cells. We examined 65 cervical neoplasms from a population of women at particularly high risk for development of carvical cancer in Xinjiang. HPV DNA analysis using type-specific PCR for the detection of HPV type 6,16 and 18. Using immunostaining for the p53 protein overexpression. Besides, seven cases unavailable for PCR, in the remaining 58 cases, 45(77.6%) cases demonstrated only positive with HPV type 16, and no one case positive with HPV type 6 or 18. In addition, using immunostaining of the p53 protein showed 49 out of 65(75.4%) positive cases. In 45 HPV positive cases, 35(77.8%) showed p53 positive, and in 13 HPV negative cases, 10(76.9%) also showed p53 positive.There was no correlation with HPV status and the p53 protein overexpression being found.
