Fas、caspase-3蛋白在颞叶癫癎病灶内的表达与神经元凋亡

摘    要:目的探讨细胞凋亡调控基因fas、caspase-3在颞叶癫癎病人病灶内的表达及意义。方法取30例 颞叶癫癎病人手术切除的病灶,应用免疫组化的方法检测凋亡相关基因fas、caspase-3的表达,同时采用光镜、电镜 及原位末端标记(TUNEL)方法检测神经细胞凋亡。结果免疫组化染色发现fas蛋白在对照组中无表达,在癫癎 组表达明显增强(P<0.001);caspase-3在对照组中有轻微表达,在癫癎组表达亦明显增强(P<0.005)。光镜检查 及TUNEL染色均未发现凋亡的神经细胞,在进行电镜检查的10例标本中有4例发现少量早期凋亡征象的神经元。 结论癫癎病人脑内存在神经元凋亡现象。fas、caspase-3基因在这一过程中可能发挥了重要的作用。

Abstract:Objective To explore the role of cell apoptosis regulatory genes of the fas and caspase-3 in the brain of human temporal lobe epilepsy. Methods The specimens were obtained from 30 patients of temporal lobe epilepsy. To detect neuronal apoptosis by microscopy, electric microscopy and TdT-mediated dUTP-biotin nick end labeling ( TUNEL) method and immunohistochemistry for fas and caspase-3. Results Unlike that in normal adult brain, fas and caspase-3 im-munoreactivity was observed in some neurons; The evidence for neuron apoplosis was not found by microscopy and TUNEL method,but some early signs of neuronal apoptosis in 4 patients(4/10) was found by electric microscopy. Conclusions These data suggest neuronal apoptosis is involved in temporal lobe of human epilepsy. Fas and caspase-3 may play a role in this process.
