丙泊酚下调miR-93-5p表达对乳腺癌MDA-MB-231细胞增殖、侵袭的影响

摘    要:目的:探讨丙泊酚对miR-93-5p以及人乳腺癌细胞MDA-MB-231增殖及侵袭的影响。方法:培养对数期MDA-MB-231细胞,进行细胞毒性试验,筛选最佳丙泊酚处理浓度与时间,实验分为空白对照组(0.1%DMSO培养基)、阳性对照组(30μmol·L^-1环磷酰胺)、丙泊酚1,2,5 mg·L^-1浓度组。CCK8法检测细胞增殖,细胞划痕及Transwell实验检测细胞迁移侵袭情况,免疫印迹法(Western Blot)检测增殖相关蛋白增殖细胞核抗原(PCNA)、侵袭迁移相关蛋白金属机制蛋白酶2(MMP-2)、金属机制蛋白酶2(MMP-9)蛋白表达情况,定量即时聚合酶链锁反应(qRT-PCR)检测细胞miR-93-5p表达情况。结果:与空白对照组相比,阳性对照组与丙泊酚各浓度组MDA-MB-231细胞增殖抑制率、迁移抑制率显著升高,裸鼠肿瘤体积、侵袭细胞数量及PCNA、MMP-2、MMP-9蛋白表达、miR-93-5p表达显著降低(P<0.05)。与阳性对照组相比,1 mg·L^-1和2 mg·L^-1丙泊酚组MDA-MB-231细胞裸鼠肿瘤体积、侵袭细胞数量及PCNA、MMP-2、MMP-9蛋白表达、miR-93-5p表达依次升高,增殖抑制率、迁移抑制率依次降低(P<0.05)。结论:丙泊酚可能通过下调miR-93-5p表达,抑制乳腺癌细胞MDA-MB-231增殖、侵袭能力发挥抗肿瘤作用。

Abstract:Objective:To investigate the effects of propofol on the proliferation and invasion of miR-93-5p and human breast cancer cell line MDA-MB-231.Methods:MDA-MB-231 cells were cultured in logarithmic phase.The optimal concentration and time of propofol treatment were selected,and the cells were divided into 5 groups:the blank control group(0.1%DMSO medium),the positive control group(30μmol·L^-1 cyclophosphamide),and propofol 1,2 and 5 mg·L^-1concentration groups.CCK8 method was used to detect the cell proliferation,cell scratch and transwell test were used to detect the cell migration and invasion,Western blotting was used to detect the expressions of proliferating cell nuclear antigen(PCNA),metalloproteinase-2(MMP-2)and metalloproteinase-9(MMP-9),and the expression of miR-93-5p was detected by qRT-PCR.Results:Compared with that of the blank control group,the inhibition rate of proliferation and migration of MDA-MB-231 cells in the positive control group and propofol group increased significantly,while the volume of tumors in nude mice,the number of invasive cells,PCNA,MMP-2,MMP-9 protein expressions and miR-93-5p expression decreased significantly(P<0.05).Compared with that of the positive control group,the tumor volume,the number of invasive cells,the expressions of PCNA,MMP-2 and MMP-9,and the expression of microRNA-93-5p in MDA-MB-231 cells of 1 mg·L^-1 and 2 mg·L^-1 propofol groups increased,while the proliferation inhibition rate and the migration inhibition rate decreased(P<0.05).Conclusion:Propofol may inhibit the proliferation and invasion of breast cancer cell MDA-MB-231 by down-regulating the expression of miR-93-5p to an anti-tumor role.
