右旋雷贝拉唑钠小鼠单次给药毒性试验研究

目的 研究单次静脉注射给予右旋雷贝拉唑钠对小鼠产生的毒性，并与左旋雷贝拉唑钠和雷贝拉唑钠进行比较。方法 采用经典半数致死量(LD₅₀)试验法进行小鼠单次给药毒性试验，动物分组后以不同的剂量给药1次，药后连续观察14 d，记录每组动物的症状反应和死亡率，观察结束剖检，观察主要脏器的病变，采用Bliss法计算半数致死量。结果 小鼠单次给予右旋雷贝拉唑钠、左旋雷贝拉唑钠和雷贝拉唑钠的半数致死量分别为192、196和193 mg·kg^-1，死亡小鼠急性毒性症状主要为挣扎、呼吸困难、俯卧不动、抽搐、角弓反张继而死亡。结论 右旋雷贝拉唑钠、左旋雷贝拉唑钠和雷贝拉唑钠单次给药后对小鼠的毒性反应和半数致死量无明显差异。

Study of single-dose toxicity of Dexrabeprazole Sodium in mice

Objective To study the single-dose toxicity of Dexrabeprazole Sodium in mice and compare the toxicity of Dexrabeprazole Sodium with Levorabeprazole Sodium and Rabeprazole Sodium.Methods The single-dose toxicities of Dexrabeprazole Sodium, Levorabeprazole Sodium and Rabeprazole Sodium in mice were evaluated with classical LD₅₀ methods.Mouse were divided into different groups and were administered with different dose for one time. The general symptoms and mortality rate were observed after administration.The animals were autopsied and lesion of main organs were observed after continuous observation for 14 days.Results The LD50 of Dexrabeprazole Sodium, Levorabeprazole Sodium and Rabeprazole Sodium in mice were 192,196,193 mg·kg^-1,respectively.The main acute toxicity symptoms of mouse were struggle,dyspnea,prone position tremble,opisthotonos and eventually to death.Conclusion The toxic reaction and LD50 of Dexrabeprazole Sodium,Levorabeprazole Sodium and Rabeprazole Sodium in mouse were almost the same.
