二参颗粒对心肌缺血大鼠血清TXA2/PGI2的平衡及炎症细胞因子水平的影响

摘    要:目的:通过检测二参颗粒对心肌缺血大鼠血清学相关指标的影响,探讨二参颗粒抗心肌缺血的机制。方法:将50只健康Wistar大鼠按随机数字表法分为5组:正常对照组、模型组、二参颗粒高、中、低剂量组(35.2,17.6,8.8 g·kg^-1),每组10只。连续灌胃给药2周后,除正常对照组外,其余4组大鼠用垂体后叶素(20 U·kg^-1)腹腔注射建立心肌缺血模型后记录心电图标Ⅱ导联变化,TUNEL染色观察大鼠心肌细胞凋亡情况,检测血清血栓素A2(TXA2)、前列环素(PGI2)的含量及炎症细胞因子白细胞介素1(IL^-1)、白细胞介素6(IL-6)和白细胞介素10(IL^-10)的水平。结果:与正常对照组相比,模型组各时间点ST段J点变化值及T波变化值、心肌细胞凋亡指数、血清TXA2含量、IL^-1及IL-6水平明显增加,PGI2含量、IL^-10水平明显降低(P<0.05)。与模型组相比,二参颗粒部分剂量组各时间点ST段J点变化值及T波变化值显著降低,3个剂量组心肌细胞凋亡指数、血清TXA2含量、IL^-1及IL-6水平显著降低,PGI2含量、IL^-10水平显著增加(P<0.05)。结论:二参颗粒能调节TXA2/PGI2的平衡,改善血清炎症细胞因子水平,从而抑制血管内皮炎症性改变,保护血管内皮功能,达到抗心肌缺血作用。

Effect of Ersheng Granules on the Balance of TXA2/PGI2 and Inflammatory Cytokine Levels in Myocardial Ischemia Rats

Abstract:Objective:To investigate the mechanism of Ershen granules against myocardial ischemia through examining the effects of Ershen granules on serological indices of myocardial ischemia rats.Methods:Totally 50 healthy Wistar rats were divided into 5 groups according to the random number table method:the normal control group,the model group,Ershen granules high,medium and low dose groups(35.2,17.6 and 8.8 g·kg^-1)with 10 rats in each group.After continuous intragastric administration for 2 weeks,except the normal control group,the other groups of rats were with intraperitoneal injection of pituitrin 20 U·kg^-1 to estblish myocardial ischemia model.The changes of electrocadiograph(ECG)Ⅱlead,apoptosis of myocardial cell,serum levels of TXA2and PGI2and serum inflammatory cytokine levels of IL^-1,IL-6 and IL^-10 were detected.Results:Compared with those in the normal control group,the variation values of J point of ST segment and T wave at each time point,the myocardial cell apoptosis index,the serum TXA2 content,and IL^-1 and IL-6 levels in the model group increased significantly,PGI2 content and IL^-10 level decreased significantly(P<0.05).Compared with those in the model group,the variation values of J point of ST segment and T wave at each time point in Ershen granules groups decreased significantly,the myocardial cell apoptosis index,the serum TXA2 content,and IL^-1 and IL-6 levels in Ershen granules groups decreased significantly,and PGI2 content and IL^-10 level increased significantly(P<0.05).Conclusion:Ershen granules can regulate the balance of TXA2/PGI2 and improve the levels of serum inflammatory cytokines,leading to inhibit the inflammatory changes of vascular endothelium,and protect the function of vascular endothelial and achieve the effect of anti-myocardial ischemia.
