莪术油中莪术醇的双波长薄层扫描测定法

摘    要:本文报道用硅胶G—CMC薄层层析,以石油醚(30~60℃)—乙酸乙酯(95:5)为展开剂,将莪术油中莪术醇与其它成分分离,用10%香草醛冰醋酸(5:1)显色,岛津CS—910双波长薄层扫描仪进行含量测定。测定采用反射法、直线扫描,测定波长λs=515nm、λR=730nm,结果由数字显示器显示。本法具有快速、准确、简便、分离效果好和微量等优点。样品显色后两小时内积分值相对稳定,取样1~7μl斑点浓度和积分值呈线性关系,回归方程y=8.30+60.97x,相关系数0.9953。含1μl以上莪术醇就可以定量,平均回收率为103.3%,同一块板上不同斑点变异系数<4.5%,同一批号不同板之间无显著性差异。

Abstract:The separation of curcumol and other substances in the valatile oil of Curcuma aromatica was accomplished on silica gel CMC plate with petroleum ether (30—60℃) and ethyl acetate (95:5)as the developer. It is treated with 10% anillin-glacial acetic acid and perchloric acid(5:1), and then determined densitometrically with a Shimadzu CS-910 dual-wavelength TLC scanner. Reflection and linear scanning were adopted with λ_s being 515nm and λ_R, being 750nm. All data were recorded and calculated by a DM-6 recorder. This method which is simple and rapid gives stable and reproducible results.
