摘    要:目的:用微生物转化的方法获得新型的黄体酮类衍生物,对于相关新药的研发至关重要。方法通过微生物转化,利用薄层层析法(TLC)和硫酸显色法跟踪监测,筛选能够有效转化黄体酮的菌株,然后通过硅胶柱层析法对发酵液中的转化产物进行分离纯化,将纯化产物进行质谱、红外光谱和核磁共振谱表征,确定转化产物的物质结构。结果本研究筛选到了1株真菌,通过分子生物学鉴定其为 Fusarium 属,该菌能够完全转化底物黄体酮,同时得到2种重要的药物中间体:雄甾-1,4-二烯-3,17-二酮和睾内酯。结论本研究筛选得到的这株菌对大规模发酵生产雄甾-1,4-二烯-3,17-二酮和睾内酯具有潜在的应用价值。

Abstract:Objective The new progesterone derivatives obtained with the microbial transformation method are vital for the research and development of new drugs.Methods TLC and Sulfuric acid methods were adopted to screen the strains which are capable of transforming progesterone with the help of microbial transformation.The silica gel column chromatography was used to purify the products in the fermented liquid,and mass spectrum, infrared spectrum and nuclear magnetic resonance spectrum were employed to identify the structure of the purified products.Results A strain which could transform progesterone completely was screened and identified as Fusarium of Molecular biology identification.Two important intermediates of hormone drugs in the purified transformation products were aquired and identified as Androst-1,4-dien-3,1 7-dione and testolactone.Conclusion The screened strain is of potential value in producing the two important intermediates of hormone drugs including Androst-1,4-dien-3,1 7-dione and testolactone in a lager scale.
