甘草酸二铵对溃疡性结肠炎大鼠的疗效及炎症因子的影响

摘    要:目的:探讨甘草酸二铵对溃疡性结肠炎(UC)大鼠的疗效及炎症因子(IFN-γ)和抗炎因子(IL-10)的影响。方法:将30只Wistar大鼠随机分成正常对照组、UC模型组和甘草酸二铵治疗组,每组10只。正常对照组大鼠自由饮用蒸馏水;UC模型组大鼠采用自由饮用葡聚糖硫酸钠(DSS)水溶液造模;甘草酸二铵治疗组大鼠在造模7d后给予甘草酸二铵灌肠治疗7d。观察各组大鼠一般症状(腹泻和肉眼血便症状)和疾病活动度(DAI积分);光学镜下观察大肠黏膜组织学的改变;通过免疫组化方法检测大鼠结肠黏膜组织中IFN-γ和IL-10的表达情况。结果:UC模型组大鼠出现腹泻及肉眼血便;甘草酸二铵治疗组大鼠仅出现不同程度腹泻,未出现明显肉眼血便;正常对照组大鼠未出现腹泻及肉眼血便情况。各组大鼠之间的DAI积分和组织学损伤评分均有统计学差异(F1=287.50,F2=125.68,P<0.05),UC模型组大鼠DAI积分和组织学损伤评分明显高于正常对照组(P<0.05),甘草酸二铵治疗组大鼠DAI积分和组织学损伤评分明显低于UC模型组(P<0.05)。病理组织学发现甘草酸二铵治疗组大鼠病理改变明显轻于UC模型组大鼠,正常对照组大鼠肠黏膜未见明显病理学改变。与正常对照组相比,UC模型组和甘草酸二铵治疗组的结肠黏膜固有层IFN-γ和IL-10表达显著增多(P<0.05);与UC模型组相比,甘草酸二铵治疗组IFN-γ的表达显著减少(P<0.05),而IL-10表达显著增多(P<0.05)。结论:甘草酸二铵能够有效减轻溃疡性结肠炎大鼠的症状(腹泻和肉眼血便)及肠黏膜病理组织学改变,其机制与调节肠黏膜炎症因子(IFN-γ)和抗炎因子(IL-10)有关。

Abstract:Objective:To evaluate the role of diammonium glycyrrhizinate(DG) in experimental colitis as well as its effect on the expression of IFN-γ and IL-10,so as to explore its underlying mechanisms.Methods:Thirty Wistar rats were randomly divided into treatment,DSS control and normal control group(n=10).The rats in treatment group were treated with DG(40 mg/kg·d).Disease activity index(DAI) and histological damage were observed.The expression of IFN-γ and IL-10 in colon tissues was detected by immunohistochemistry.Results:Compared with the DSS control group,the DAI and histological injury score of colon tissue in treatment group were significantly improved(F1=287.50,F2=125.68,P<0.05),and the expression of IFN-γ in treatment group decreased significantly(P<0.05),and the expression of IL-10 in treatment group increased significantly(P<0.05).Conclusion:Our study indicates that DG can relieve colitis in rats induced by DSS.By affecting the expression of IFN-γ and IL-10 to inhibit inflammation may be one of the possible mechanisms.
