川芎嗪对癫痫模型海马内信号分子的影响

摘    要:目的:研究戊四氮(pentylenetetrazol,PTZ)点燃癫痫模型大鼠海马内环磷腺苷(cyclic adenosine monophos-phate,cAMP)、环-磷酸鸟苷(cyclic guanosine monophosphate,cGMP)的变化及川芎嗪(Tetramethylpyrazine,TMP)对其的影响。方法:将实验动物随机分为3组,正常对照组、癫痫模型组、TMP治疗组。采用放射免疫法测定海马内cAMP、cGMP的含量。结果:模型组与常对照组比,cAMP、cGMP含量明显增加,cAMP/cGMP比值明显减小,差异均具有显著性意义(P<0.01),TMP治疗组与癫痫模型组比,cAMP含量明显增加,cGMP含量则明显减少,cAMP/cGMP比值明显增加,差异均具有显著性意义(P<0.01)。结论:川芎嗪抑制癫痫放电作用与其调节海马内cAMP、cGMP含量变化有关。

Abstract:Objective:To study the effect of tetramethylpyrazine(TMP) in treating epilepsy and its relation to signaling moleculein hippocampusof rats.Methods:Experimental animals were selected and randomized into three groups:normal saline control group,pentylenetetrazol group and treatment of tetramethylpyrazine group,and the changes of the content of cAMP and cGMP in hippocampus of rats were detected using radioimmunity method.Results: The content of the cAMP and cGMP in hippocampus in pentylenetetrazol groups were significantly higher than in thosesaline control groups.While the proportionality in pentylenetetrazol groups were significantly lower than in saline control groups(P<0.05).The content of cAMP in hippocampusin those in treatment of tetramethylpyrazine groups were significantly higher than those in pentylenetetrazol groups.While the contentof the cGMP in hippocampusin in treatment of tetramethylpyrazine groups were significantly lower than in those pentylenetetrazol groups(P<0.05).The proportionality in pentylenetetrazolgroups were significantly higher than in saline control groups(P<0.05).Conclusion:Tetramethylpyrazine inhibit epilepsy discharge may be its regulationthe content of cAMP and cGMP in hippocampus.
