黄芪对低氧性肺动脉高压大鼠肺组织血栓素A_2及肺血管结构的影响

摘    要:目的:探讨黄芪对低氧性肺动脉高压(HPH)大鼠肺腺泡内肺动脉结构重建及肺组织血栓素(TXA2)的影响。方法:雄性Wistar大鼠21只,随机分为3组,每组7只。对照组:室内空气正常饲养;低氧组:在常压低氧舱内(10±0.5)%饲养;治疗组:缺氧+黄芪(12g/kg)。低氧30天分别用右心导管法测定肺动脉平均压(mPAP),放射免疫法测定肺组织中TXA2含量,计算MA、PMA、NMA比例及肺中、小肌型动脉相对中膜厚度(RMT)与相对中膜面积(RMA)。结果:低氧组mPAP、RVHI、TXA2含量较对照组明显升高(P<0.01或P<0.05),治疗组较低氧组明显减少(P<0.05)。低氧组肺小动脉肌化程度、RMT(%)、RMA(%)较对照组明显增多(P<0.01),治疗组较低氧组明显减少(P<0.01)。结论:慢性低氧可引起大鼠肺血管结构重建,黄芪注射液可降低肺组织TXA2含量,可部分逆转肺血管结构重建,降低肺动脉高压。

Abstract:Objective:To study the influence of Radix Astragali on structural remodeling of intra-acinar pulmonary artery(IAPA) and Pulmonary tissue TXA2 in hypoxic pulmonary hypertension rats.Methods:21 male Wistar rats were divided into three group at random,Control group(C);hypoxia group(H);hypoxia Radix Astragali group(T).Rats in T group were treated with an intraperitional injection of Radix Astragali(12g/kg).MPAP,mCAP,RVHI,TXA2 were measured.Muscularizing degree of IAPA and RMT(relative medial thickness),RMA(relative medial acreage) of intra-acinar pulmonary mucularized artery were also measured.Results:MPAP,RVHI,TXA2 in T group were significantly lower than that in H group(P<0.05).The rate of muscle artery(MA),partially muscle artery(PMA),RMT and RMA in T group were much lower than that in H group(P<0.01).Conclusion: Radix Astragali could regulate TXA2 and reversed the remodeling of Intra-acinar pulmonary artery partly.
