摘    要: 目的 建立三七总皂苷和三七剪口的高效液相色谱(HPLC)指纹图谱和含量测定方法,为三七总皂苷的质量控制提供有效手段。 方法 采用三七总皂苷对照品作为随行对照, 用岛津 C18(150 mm × 4.6 mm,5 μm)色谱柱,乙腈-水梯度洗脱,检测波长 203 nm,流速均为 1.0 ml·min-1,确定构成三七总皂苷指纹的 10 个共有峰。 结果 采用梯度洗脱的方法能使三七总皂苷的 5 种主要成分三七皂苷 R1、人参皂苷 Rg1、Re、Rb1 和 Rd 实现基线分离,并可对这 5 种主要成分进行定量。以人参皂苷 Rgl 为参比峰,所测定的 10 批三七总皂苷样品的相似度均 ≥ 0.995. 结论 所建立的 HPLC 指纹图谱和测定方法以三七总皂苷对照品作为随行对照,可避免由于仪器、试剂和人员的不同所产生的误差,并且简化了实验操作,可作为三七总皂苷的质量控制手段。

HPLC fingerprints of rhizome of Panax notoginseng and notoginsenosides
Abstract:Objective To establish the HPLC fingerprints of rhizome of Panax notoginseng and notoginsenosides, and find a method to determine their contents, so that to control the quality of production of notoginsenosides. Methods The reference substance of notoginsenosides was used as the accompany reference. HPLC was carried on Shimadzu C18 column (250 mm × 4.6 mm, 5 μm) with mixtures of acetonitrile and water as the mobile phase in a gradient mode.The detection wavelength was set at 203 nm and the flow rate was 1.0 ml·min-1. Totally 10 common peaks were identified as the characteristic fingerprints of notoginsenosides. Results By using gradient mode, the 5 major components of notoginsenosides, notoginsenosides R1, and ginsenosides Rg1, Re, Rb1, and Rd were separated completely, and their quantity could be determinated. With the ginsenosides Rg1 as the reference peak, the 10 samples exhibited high similarities (over 0.995). Conclusion By using notoginsenosides as the reference substance, the instruments, reagents, and manipulation-induced errors can be avoided. Moreover, because the method is simple, it can be used for qulity control.
