Abstract:Objective To investigate the role of Sestrin 2 in epithelial mesenchymal transition (EMT) of pancreatic cancer PANC-1 and CFPAC-1. Methods The expression of Sestrin 2 in different pancreatic cancer cells was intervened. The changes of EMT were detected by wound healing assay, Transwell invasion assay, RT-PCR, Western blotting and animal test. Meanwhile, the expression of Sestrin 2 in human body was analyzed.Results In the Transwell invasion assay and the wound healing assay, the migration ability and invasion ability of pancreatic cancer cells with high expression of Sestrin 2 were significantly decreased (P<0.05); In the animal test, the tumor growth of pancreatic cancer cells with high expression of Sestrin 2 was significantly decreased (P<0.05); In the RT-PCR and Western blotting analysis, the expression level of E-cadherin was significantly increased (P<0.05), while the expression levels of N-Cadherin and Vimentin were significantly decreased (P<0.05) in the pancreatic cancer cells with high expression of Sestrin 2. The expression of Sestrin 2 was low in human pancreatic cancer; with the increase of Sestrin 2 expression, the expression level of p60-s6k, a key molecule in mTOR signaling pathway, decreased significantly (P<0.05). Conclusion This study suggests that Sestrin 2 can inhibit the EMT of pancreatic cancer, which playing a role through mTOR signaling pathway.
