PAQR4在肝细胞癌组织中的表达及其在细胞增殖、迁移和侵袭中的作用

目的 探讨孕激素和脂联素受体4（PAQR4）在肝细胞癌（HCC）中起到的调控作用。方法 （1）在TCGA数据库中调查HCC组织中PAQR4的表达，分析PAQR4与HCC患者预后的关系。（2）转染两种不同的PAQR4特异性siRNA致HCC细胞系Hep3B和Huh7中PAQR4的表达降低，通过细胞增殖、侵袭和迁移实验来分析PAQR4对HCC的影响。（3）通过TCGA获取数据并采用Pearson卡方检验评估PAQR4的表达与HCC组织中免疫细胞浸润的关系。结果 （1）HCC患者肝脏肿瘤组织中PAQR4表达水平高于癌旁组织（P＜0.05），PAQR4高表达与HCC患者预后不良相关（P=0.0014）。（2）在Hep3B和Huh7细胞中敲低PAQR4可抑制细胞增殖、侵袭和迁移。（3）PAQR4在HCC组织中的高表达与多种免疫细胞浸润存在相关。结论 PAQR4与肝细胞癌具有相关性，敲低PAQR4会抑制HCC细胞的增殖、侵袭和迁移。

Expression of PAQR4 in hepatocellular carcinoma tissues and its role in cell proliferation,invasion and migration

Objective To explore the regulatory role of progestin and adipoQ receptor 4 (PAQR4) in hepatocellular carcinoma (HCC). Methods (1) To investigate the expression of PAQR4 in hepatocellular carcinoma tissues in TCGA database, and to analyze the relationship between PAQR4 expression and of HCC prognosis. (2) Down-rugulation of PAQR4 in HCC cell lines Hep3B and Huh7 by transfection two different PAQR4 specific siRNA. Effect of PAQR4 on HCC was investigated by analyzing the cell proliferation, invasion and migration in HCC cells. (3) Pearson chi-square test was used to valuate the relationship between the expression of PAQR4 and the infiltration of immune cells in HCC in TCGA database. Results (1)The expression level of PAQR4 in HCC tissues was higher than that in paracancerous tissue (P<0.05). High expression level of PAQR4 was related with the poor prognosis of HCC (P=0.0014). (2) Knockdown of PAQR4 could inhibit the proliferation, invasion and migration in HCC cell lines Hep3B and Huh7. (3) High expression of PAQR4 in HCC paracancerious tissue was involved in the infiltration of various immune cells. Conclusion PAQR4 is associated with HCC. Knockdown of PAQR4 can inhibit the cell proliferation, invasion and migration in HCC cell line.
