地灵丹对大鼠高尿酸血症肾病影响的实验研究

摘    要:目的观察地灵丹对大鼠高尿酸血症肾病的影响,并探讨相关作用机制。方法采用高嘌呤饲料制作高尿酸血症肾病大鼠模型,分为地灵丹治疗组、别嘌呤醇组、模型对照组及正常对照组,每组15只大鼠,分别给予相应的干预措施,持续4周。检测大鼠血尿酸、肌酐变化,观察肾脏组织学变化及细胞因子TGF-β1、TNF-α的表达。结果实验第14天,与正常对照组比较,各造模组血尿酸、血肌酐水平显著升高,差异有统计学意义(P〈0.05,P〈0.01);与模型组比较,地灵丹组及别嘌呤醇组血尿酸、血肌酐水平均显著降低,差异有统计学意义(P〈0.01)。实验第28天,与正常对照组比较,模型组及地灵丹组血尿酸、血肌酐水平显著升高,差异有统计学意义(P〈0.01),而别嘌呤醇组血尿酸水平出现显著降低,差异有统计学意义(P〈0.01);与模型组比较,地灵丹组及别嘌呤醇组血尿酸、血肌酐水平显著降低,差异有统计学意义(P〈0.01);与地灵丹组比较,别嘌呤醇组的血尿酸水平显著降低,血肌酐水平显著升高,差异有统计学意义(P〈0.01);与正常对照组比较,其余各组TGF-β1、TNF-α阳性面积均显著增多,差异有统计学意义(P〈0.01);与模型组比较,地灵丹组与别嘌呤醇组的TGF-β1阳性面积显著减少,差异有统计学意义(P〈0.05,P〈0.01)。与模型组比较,地灵丹组与别嘌呤醇组大鼠肾脏病理改变减轻。结论地灵丹可减缓高尿酸血症肾病大鼠肾脏损害的作用,其作用机制可能与调节TGF-β1、TNF-α等细胞因子有关。
