摘    要:目的 使用3种培养液对海马神经元进行培养,遴选出一种使神经元存活率较高、存活时间较长的培养液。方法 分离SD大鼠胎鼠的海马神经元,分别用DMEM、DMEM/F12和Neurobasal培养液进行培养,应用倒置相差显微镜、台盼蓝排斥实验、MTT法和LDH释放率法进行细胞形态和细胞活力分析。结果 Neurobasal培养液培养的神经元细胞活力,与另二组培养液培养的神经元活力相比差异有显著性(P〈0.05)。结论 Neurobasal培养液可作为海马神经元体外培养的良好培养液。

Abstract:Objective To determine an optimal nutrient medium from three sorts of nutrient medium, in which the hippocampal neurons have the highest survival rate and longest survival time. Methods Hippocampal neurons were dissected from SD foetal rats and plated in DMEM, DMEM/F12 and Neurobasal nutrient medium respectively. The neuronal morphology was visualized using inverted phase-contrast microscope and trypan blue exclusion. MTT assays and the LDH release were used to evaluate the neurons viability. Results The viability of cells in Neurobasal nutrient medium is significantly different from that of the others (P<0.05). Conclusion Neurobasal nutrient is the best medium for culturing primary hippocampal neurons.
