西瑞香素对人肝癌HepG2细胞增殖、凋亡及细胞周期的影响

摘    要:目的 探讨西瑞香素对人肝癌HepG2细胞增殖、凋亡及细胞周期的影响.方法 用不同浓度的西瑞香素作用于体外培养的HepG2细胞,采用四甲基偶氮唑蓝法(MTT)检测西瑞香素对HepG2细胞增殖抑制能力;采用AnnexinV/PI双染流式细胞术测定西瑞香素对HepG2细胞凋亡的影响;采用PI单染流式细胞术检测西瑞香素对HepG2细胞周期的影响.结果 西瑞香素可抑制HepG2细胞的增殖,且存在明显的浓度和时间依赖关系;流式细胞术检测显示西瑞香素作用48 h后,实验组细胞未出现明显的早期凋亡细胞群.但实验组处于S期的细胞数较对照组明显减少(P<0.05),G0/G1期的细胞较对照组明显增多(P<0.05).结论 西瑞香素对人肝癌HepG2细胞的体外增殖具有抑制作用,可能与阻滞细胞周期有关.

Abstract:Objective To observe the effects of daphnoretin on the proliferation, apoptosis and cell cycle of human hepatoma HepG2 cells. Methods The human HepG2 cells were cultured and treated by daphnoretin in different concentrations in vitro. The inhibitive effects on the cell growth were tested using Monotetrazdium (MTT) assay. The apoptosis and cell cycle were determined by flow cytometry analysis. Results Daphnoretin had inhibitive effects on the growth of HepG2 cells in a concentration and time-dependent manner. Compared with the control, the percentage of early apoptotic rate in daphnoretin treatment group was not higher, but the number of cells in S phase was reduced significantly (P〈0.05) and the number of cells in G0/G1 phase was increased (P〈0.05). Conclusion It is suggested that daphnoretin could inhibit the proliferation of HepG2 cells, which is associated with the arrest of cell cycle.
