摘    要:用酶消化法自人脐带静脉分离内皮细胞,进行体外传代培养。观察EHI病毒在该细胞中的增殖和复制。结果表明,从第一代培养第7日起,在感染内皮细胞胞浆内可检出EHF病毒抗原;阳性细胞数及胞浆内病毒抗原含量随培养时间的延长和传代次数的增加而增多,而感染细胞未见明显病变。提示EHF病毒可侵入人血管内皮细胞增殖复制,参与EHF的发病过程。

Abstract:The infection of epidemic haemorrhagic fever virus (EHFV) in human endothelial cells (HEC) in vitro was studied. The culture of HEC was established with human umbilical vein cells isolated by collagenase digestion, into which the 76-118 strain of Hantaan virus was inoculated. The results described here showed that EHFV antigen could be detected in the cytoplasm' of the infected HEC on the seventh day of the first passage by the immunoflurorescent technique, and that the increments of the detectable positive cells and viral antigens in He cytoplasm of the infected cells were consistent with the prolongation of culture and the frequency of passages. It was observed that EHFV infection in HEC in vitro induced no apparent cytopathogenic effects. It is suggested that the HEC may be the target cells susceptible to EHFV infection and that the replication of EHFV in HEC may be an important initiating factor in the pathogenesis of the disease.
