山茱萸环烯醚萜总苷对糖尿病大鼠肾形态学及其Na+,K+-ATP酶活性的影响

摘    要:目的:从形态学及蛋白酶的角度探讨山茱萸环烯醚萜总苷(ICO)防治糖尿病肾病(DN)的作用。方法:腹腔注射链脲佐菌素造成糖尿病大鼠模型,将造模成功的大鼠按血糖值随机分为模型组、氨基胍组和ICO组,同期另没一正常组,各组连续灌胃12周。实验结束后用光镜观察肾形态学变化,试剂盒测定肾组织细胞膜Na^+,K^+-ATP酶的活性。结果:模型组大鼠肾脏光镜检查肾小球和肾小管病变明显。而且肾组织细胞膜Na^+,K^+-ATP酶活性低于正常组。而ICO组病变明显减轻,表现出较好的抗肾小球内基质增生、抗肾小管坏死和改善Na^+,K^+-ATP酶活性等作用。结论:ICO可改善糖尿病大鼠肾组织形态学病变及修复Na^+,K^+-ATP酶活性,对DN所引起的结构和功能改变都具有一定的防治作用。

Abstract:Aim To study the protective effect of iridosides of cornus officinalis(ICO)on diabetic nephropathy(DN)from the renal morphology and protease.Methods:Diabetic rats were induced by ip.Streptozotoin,The model rats were divided randomly into three groups:control,aminoguanidine and ICO.One more normal group was added.Those rats were fed with each drug for twelve weeks.The morphology of kidney were observed under light microscope.The Na~+,K~+-ATPase activity were tested by Na~+,K~+-ATPase test Kit.Result:The model group showed apparent pathological changes in glomerulus and renal tubule.The Na~+,K~+-ATPase activity in model groups remarkably decreased.While ICO can relieve mesangial matrix hyperplasia,prevent renal tubular from necrosis and recover Na~+,K~+-ATPase activity.Conclusion:From the ameliorative role of ICO on renal morphology and Na~+,K~+-ATPase activity,we conclude that it should be beneficial to the prevention of structural and functional damage of the diabetic nephropathy.
