高压氧对海水淹溺肺水肿兔肺c-fos和c-jun基因表达的影响

摘    要:目的 观察高压氧对海水淹溺肺水肿(PE-SWD)兔肺c-fos,c-jun基因表达的影响并探讨其作用机制。方法 制备PE-SWD动物模型,分别采用高频通气、联合用药治疗和高频通气、联合用药、高压氧(HBO)治疗。用原位杂交法观察3组兔肺内c-fos和c-jun基因水平。结果 对照组肺泡上皮细胞和毛细血管内皮细胞胞浆中有大量c-fos和c-jun mRNA。用药治疗组和HBO组兔肺内c-fos和c-jun基因表达较对照组明显减少,尤以HBO4组减少更甚。结论 HBO可改善受损肺组织氧气供给,减轻其损伤,从而减少c-fos,c-jun的表达。

Abstract:Objective To study the effects of hyperbaric oxygen(HBO) on the expression of c-fos and c-jun genes in pulmonary cells of rabbit with pulmonary edema after sea-water drowning(PE-SWD) and its action mechanisms. Methods PE-SWD animals were induced,and divided into three groups:PE-SWD group(control group),medicine group[medicines high frequency jet ventilation(HFJV)]and HBO group(medicines HFJV HBO).Expression of c-fos and c-jun genes were detected by the method of hybridization in situ. Results There were a lot of c-fos mRNA and c-jun mRNA in the alveolar epithelial cells and capillary endothelial cells of control group,whereas the expression of c-fos and c-jun genes of either medicine group or HBO group decreased obviously,especially of the latter. Conclusions HBO could improve the O 2 supply,reduce the cells' damage,thus the expression of the pulmonary cells' c-fos and c-jun genes would be decreased.
