骨连结蛋白在大鼠正常牙髓和牙髓修复中的基因表达

摘    要:目的 :利用原位杂交技术 ,观察骨连结蛋白 (osteonectin ,ON)在大鼠正常牙髓和牙髓修复过程中的基因表达特征。方法 :在Wistar大鼠上下颌第一磨牙近中面制备一单面洞。分别观察 3d和 15d后处死动物。组织学处理后切片。体外转录合成单链RNA探针并标记地高辛 (Digoxigenin)。将标记探针与标本进行原位杂交。利用生物素标记的抗地高辛抗体和SABC标记 ,DAB显示杂交信号。苏木素复染。结果 :正常牙髓组织中 ,ON基因表达于牙髓成牙本质细胞、牙髓成纤维细胞内。ON在备洞 3d后的成牙本质细胞和成牙本质细胞层下方的牙髓细胞内表达强阳性。 15d后 ,可见修复性牙本质形成。修复性牙本质下方的成牙本质细胞样细胞表达ON。结论 :在牙髓修复过程中 ,ON在牙髓修复过程中通过自分泌途径 ,参与了修复性牙本质的形成和矿化

Abstract:Objective: To investigate the gene expression of ON (osteonectin) in normal dental pulp and repairing pulp of rats. Methods: Simple cavities were prepared in the mesial surface of the first molars of both maxilla and mandible. The animals were sacrificed in 3 and 15 days of post operation respectively. After histological process, the paraffin sections were hybridized with ON cRNA probe. Results: ON mRNA was expressed in normal dental pulp cells(odontoblasts and dental pulp fibroblasts) in the second molar. The strong hybridization signals were observed in odontoblasts under the cavity in "3 days" group. In "15 days" group, ON was expressed in reparative dentin formed odontoblast like cells. Conclusion: ON is involved in the reparative dentinogenesis as a non mineralization specific protein.
