脑胶质瘤PS值与MMP-2和MMP-9表达相关性的实验研究

摘    要:目的 运用CT灌注成像评价大鼠C6脑胶质瘤生长过程中血管通透性的变化及其与MMP-2、MMP-9表达的相关性.方法 采用立体定向方法在30只雌性Wistar大鼠右侧尾状核接种C6细胞,于接种后1 W、2 W、3 W分别进行CT灌注成像,每次检查结束后留取鼠脑标本作病理学检查.结果 各组肿瘤区PS值均比对侧正常脑组织PS值增大(P<0.01),荷瘤2 W及3 W组肿瘤中心区PS值比相应肿瘤周边区PS值增大(P<0.01),与荷瘤1 W组比较,荷瘤2 W和3 W组肿瘤中心、肿瘤周边区PS值增大(P<0.05);肿瘤区MMP-2和MMP-9阳性表达随肿瘤生长而增加;肿瘤周边区PS值与MMP-9表达呈正相关(rx=0.540,P<0.05).结论 PS值可反映大鼠C6脑胶质瘤生长过程中肿瘤不同区域血管通透性的动态变化,肿瘤周边区域PS值变化与MMP-9表达密切相关.

Abstract:Objective To evaluate changes of permeability surface (PS) of rat C6 brain gliomas by computed tomography perfusion and to study correlation between PS and expression of MMP-2 and MMP-9. Methods C6 cells were inoculated into the right caudate nucleus of 30 Wistar rats by stereotactic procedure to established animal model. CT perfusion was performed on 1 week, 2 and 3 weeks after implantation. Immediately after each examination, some rats were sacrificed for pathology. Results All PS values of the tumors at 1 week, 2 weeks and 3 weeks after implantation increased compared to those of the contralateral sides (P<0.01). The PS values in the center of the tumors at 2 and 3 weeks after implantation were more than that in the peripheral regions of the tumors (P<0.01). Statistical difference of PS values was present comparing with those of the tumors at 1 week and both central and peripheral tumoral regions of 2 weeks and 3 weeks after implantation (P<0.05). Expression of MMP-2 and MMP-9 were observed adjacent to the vascular especially at the periphery of the tumors, and positive correlation was present between PS and expression of MMP-9 under the development of the tumors (rs=0.540, P<0.05 ). Conclusion The PS can display permeability of different tumor regions with the development of rat C6 brain gliomas in vivo, and close correlation may exists between expression of MMP-9 and PS values.
