肺癌平药物血清对A549人肺癌细胞凋亡及基因表达的影响

摘    要:目的探讨肺癌平治疗肺癌的作用机制。方法采用血清药理学方法孵育A549人肺癌细胞,通过电镜、流式细胞术、免疫细胞化学方法检测肺癌平药物血清作用后对细胞凋亡及相关基因bcl-2、p53表达的影响。结果肺癌平药物血清作用后电镜下可见凋亡小体形成,流式细胞仪检测可见亚二倍体核型峰的特征,免疫细胞化学法显示抑癌基因p53主要定位在细胞核,部分细胞浆也有淡染。癌基因bcl-2主要表现为胞浆着色,流式细胞仪分析药物血清作用后,p53表达增强,bcl-2表达降低,随着作用时间延长,两种基因表达明显增强。结论肺癌平治疗肺癌的作用机制是通过诱导肺癌细胞凋亡及影响相关基因bcl-2、p53表达实现的。

Abstract:Objective To discuss the mechanism of Feiaiping contained serum in anti-tumor action. Methods Using serum pharmacological method to culture human lung cancer cell line A_~549 , the effect of Feiaiping contained serum on cell apoptosis and the related gene expressions of bcl-2 and p53 were observed by electron microscope (EM), flow cytometry assay and immunocytochemistry. Results After treated with Feiaiping contained serum, apoptosis bodies were observed under EM, hypodiploid caryotype peak was observed by flow cytometry assay. Immunocytochemistry indicated that anti-oncogene p53 mainly localized in nucleus with partial light staining cytoplasm. Oncogene bcl-2 was stained in cytoplasm. The expression of p53 increased after flow cytometry assayed, while the expression of bcl-2 decreased. Both of gene expressions of bcl-2 and p53 increased obviously along with the time prolonged. Conclusions The mechanism of Feiaiping in anti-tumor might be achieved through inducing apoptosis and work on the related gene expression of bcl-2 and p53.
