摘    要:目的研究长时连续作业对大鼠促觉醒肽orexin- A及受体表达的影响。方法使用长时连续作业动物模型,采用免疫组织化学方法结合图像分析技术测定orexin -A及受体OX1R在下丘脑外侧区,背缝核的变化。结果经过长时连续作业训练后,跑台组与对照组相比,orexin- A阳性细胞在下丘脑外侧区、orexin- A阳性纤维在背缝核及OX1R在背缝核均显著减少。结论长时连续作业可造成促觉醒肽orexin- A和OX1R的表达降低。

Abstract:Objective To investigate the effects of long-term continuous operations on orexin-A and its receptor (OX1R) expression in the lateral hypothalamic area (LHA) and dorsal raphe nucleus (DR) of a rat model. Methods Rats exercised for two circadian cycles using a treadmill protocol. Subsequently, immunohistochemical techniques combined with quantitative analysis of images techniques were performed to detect the expression of orexin-A and OX1R in rat LHA and DR. Results The number of orexin-A immunoreactive neurons in LHA, orexin-A immunoreactive fibers and OX1R in DR of operational animals was significantly decreased as compared with control group (P<0.01). Conclusion Rat alertness activities are deleteriously influenced by long-term continuous operations, which may be related to the depressed expression of orexin-A and its receptor in the arousal systems.
