激光透穴离子导入法对局灶性脑缺血再灌注大鼠脑组织NO和NOS的影响

摘    要:目的:观察激光透穴离子导入法对大鼠大脑中动脉缺血再灌注损伤的影响。方法:SD大鼠40只随机分为4组,即假手术组、模型组、电针组、激光透穴离子导入组。采用线栓法制备大鼠大脑中动脉缺血再灌注模型,电针组和激光透穴离子导入组在造模后即刻、16h分别进行治疗,并观察各组的左右两侧大脑半球皮层含水量的变化,并检测右侧海马中的一氧化氮(NO)的含量及一氧化氮合酶(NOS)的活性。结果:激光透穴离子导入组的缺血侧脑含水量下降,NO含量明显减少,NOS活性下降。结论:激光透穴离子导入法可下调脑缺血再灌注后NOS的表达,减少NO的生成,以保护脑缺血再灌注损伤。

Abstract:Objective: To investigate the effects of laser irradiation and herbal iontophoresis at acupoints against brain damage after middle cerebral artery occlusion(MCAO) in rats.Methods: Forty SD rats were randomly divided into four groups,including sham group,model group,electro-acupuncture group and laser irradiation group.The cerebral ischemia-reperfusion(I/R) model was established by thread embolism of middle cerebral artery.The rats in laser group,as well as the electro-acupuncture group were given treatment as soon as the occlusion finished and 16 hours after the occlusion.The brain water content of cortexes at both cerebral hemispheres was observed.The content of nitric oxide(NO) and the activity of nitric oxide synthase(NOS) in the right hippocampi were detected.Results: The brain water content of laser group was lower than that of the model group;the content of NO and the activity of NOS decreased significantly after treatment.Conclusion: This method can prevent focal cerebral ischemia-reperfusion injury possibly by suppressing NOS expression and decreasing NO production.
