HPLC法测定大黄配方颗粒中大黄酸、大黄素、大黄酚的含量

摘    要:目的建立大黄配方颗粒中大黄酸、大黄素、大黄酚的含量测定方法。方法采用反相高效液相色谱法测定,用Kromasil ODS柱(250 mm×4.6 mm,5μm),以甲醇-0.1%磷酸溶液(85∶15)为流动相,检测波长254 nm。结果大黄酸、大黄素、大黄酚的线性范围分别为0.010 9~0.218 0μg、0.011 2~0.224 0μg和0.011 9~0.238 0μg;大黄酸平均回收率为100.6%,RSD=0.92%(n=9);大黄素平均回收率为100.5%,RSD=0.85%(n=9);大黄酚平均回收率为100.6%,RSD=1.15%(n=9)。结论所用方法测定样品分离效果佳,灵敏度高、重复性好,能有效地控制大黄配方颗粒的质量。

Abstract:Objective To establish a quantitative method for rhein,emodin and chrysophanol in Dahuang Dispensing Granule by RP-HPLC.Methods The sample were separated by Kromasil ODS column(250 mm×4.6 mm,5 μm)with methanol-phosphoric acid 0.1%(85∶15)as mobile phase,detection wavelength at UV 254 nm.Results The linear relationship was the range of rhein within 0.010 9~0.218 0 μg,emodin in 0.011 2~0.224 0 μg and chrysophanol in 0.011 9~0.238 0 μg,respectively.The average recovery of rhein was 100.6% with RSD of 0.92%(n=9),emodin was 100.5%with RSD of 0.85%(n=9) and chrysophanol was 100.6% with RSD of 1.15%(n=9),respectively.Conclusion The method is available with a good separable efficiency,reproducibility and sensitivity,which can be well controlled the quality of Dahuang Dispensing Granule.
