硫酸镁对血栓形成及血小板活化/聚集的影响

摘    要:目的 复制FeCl3损伤血管内皮诱导大鼠颈总动脉血栓形成模型,探讨血栓形成时血小板活化和聚集变化及硫酸镁对其影响。方法 将SD大鼠40只随机分为4组,即假血栓对照组、单纯血栓组,此两组均给予生理盐水;Mg-early组和Mg-late组,此两组分别在应用FeCl3前10min和后7min开始持续给予MgSO4,给药1h后颈总动脉取血测定血小板聚集率和血小板表面a颗粒膜蛋白140,血栓段脉管病理形态学检查。结果 早期给镁组明显抑制血栓形成,减轻血栓段脉管重量,抑制血小板聚集,降低血浆GMP-140含量,晚期给镁组对血栓形成及上述指标则无明显影响。结论 血栓形成过程存在血小板活化、聚集,早期给予MgSO4可能抑制血栓形成和血小板活化、聚集。

Abstract:Objective The change of platelet activation/aggregation and the effect of magnesium sulfate were discovered in the rat model with carotid thrombosis induced by FeCl 3 , which damaged vascular endothelium. Methods Forty rats were randomly divided into four groups. The rats in normal group and thrombosis group were given 0.9% sodium chloride. The rats in Mg-early group and Mg-late group were given magnesium sulfate before the topical application of FeCl 3 ten minutes and after it seven minutes. After one hour administration, the platelet aggregation and the number of alpha-granule membrane protein-140 (GMP-140) molecule on platelet surface were measured. The vessels with thrombus were made morphological.Results Mg-early group inhibited thrombosis, lightened the weight of thrombus, inhibited platelet aggregation and reduced the concentration of GMP-140. Mg-late group had no distinctive effect. Conclusions Early administration of intravenous magnesium sulfate may inhibit thrombosis and platelet aggregation/activation.
