摘    要:背景神经元的功能状态与机体的生命活动密切相关.神经元受损后生长相关蛋白-43(GAP-43)表达预示着神经元是否有再生.目的探讨施万细胞移植对损伤的大鼠中脑神经元修复的影响.设计随机对照实验研究.地点和对象实验地点为北京市神经外科研究所;研究对象Wistar大鼠,雌雄不限,体质量(180±20)g,随机分为实验组和对照组,每组6只动物.干预BrdU标记的新生大鼠施万细胞移植至电针损伤的大鼠中脑区域,不同时间处死动物,免疫组织化学方法观察BrdU和GAP-43的表达并通过图像分析系统处理实验结果.主要观察指标移植的施万细胞在脑内的存活时间;损伤神经元的再生情况.结果施万细胞移植后8个月仍可见BrdU阳性细胞,其数目增加15%,并主要向大脑皮质迁移;移植后1个月损伤的中脑神经元GAP-43的表达与对照组相比差异有显著性意义.结论施万细胞移植可促进中脑损伤神经元的修复.

Abstract:BACKGROUND: Activities of the human body are closely related to neuron function; Expression of growth associated protein-43(GAP-43) in neuron represents the regeneration of injured neurons.OBJECTIVE: To explore the effects of Schwann cell transplantation on the plerosis of injured midbrain neuron.DESIGN: A randomized controlled trial study.SETTING and MATERIALS: Setting: Beijing Neurosurgical Institute.Materials: Wistar rats of either sex, weighting( 180 ± 20) g, were divided into experimental group and control group with 6 in each.INTERVENTION: Schwann cells of neonatal rats, which were labeled by BrdU, were transplanted into rat midbrain area prior to injury with electric needle stimulus. The rats were killed at different time points. Immunohistochemistry and color imaging analyzer were used to explore the expression of BrdU and GAP-43 as well as quantitative changes.MAIN OUTCOME MEASURES: Survival time of transplanted Schwann cells in the brain and regeneration condition of injured neuron.RESULTS: BrdU positive cells could be identified for up to 8 months of Schwann cell transplantation and the number increased by 15%. The cells mainly migrated toward the injured ipsilateral cortex. The expression of GAP-43 of injured midbrain neuron(0. 533 ±0.005) one month after the transplantation significantly differed from that of control group(0. 121 ±0.007)(t=2.589, P <0.05).CONCLUSION: The transplantation of Schwann cells can promote the plerosis of injured midbrain neuron.
