基于活体成像技术的裸鼠卵巢癌休眠模型的建立

摘    要:目的 建立基于活体成像的裸鼠卵巢癌休眠模型并利用小动物活体成像技术验证休眠细胞的存在.方法 采用慢病毒载体对人卵巢癌skov-3细胞稳定转染萤火虫荧光素酶基因(firefly luciferase gene),经筛选获得稳定表达荧光素酶基因的人卵巢癌细胞株(skov-3/luc).将skov-3/luc细胞接种至裸小鼠右侧腋部皮下,成瘤后观测荧光并做肿瘤切除手术,术后的裸鼠再常规喂养8周后选取接种肿瘤处肉眼未见肿瘤组织生长的裸鼠进行活体成像观察,显示接种处肿瘤细胞荧光阳性者,即认为卵巢癌裸鼠休眠模型建立成功.结果 筛选到了稳定表达Luc的skov-3/luc细胞株;成功构建了基于活体成像的裸鼠卵巢癌休眠模型,并利用小动物活体成像技术验证了裸鼠体内卵巢癌休眠细胞的存在.结论 利用活体成像技术构建的裸鼠卵巢癌休眠模型为研究肿瘤休眠的机制及诱导和维持肿瘤细胞休眠的方法提供了理想的实验动物模型.

Abstract:Objective To establish ovarian cancer dormancy of nude mouse model based on the in vivo imaging technology, and use the small animals living imaging techniques to verify the existence of the dormant cells.Methods human ovarian cancer skov 3 cells were transfected with firefly luciferase gene via slow virus vector.After that the skov 3 cells with stable expression of firefly luciferase gene (skov-3/luc) were obtained by screening.The skov-3/luc cells were selected and inoculated into nude mice of right axillary region under the skin.The skin tumor was observed on fluorescent and subsequently resected.The nude mice were fed for 8 weeks after the operation, the nude mice with no gross tumor were selected for further observation with fluorescent by ling imaging.The dormant ovarian cancer nude mouse model was established with positive in vivo imaging.Results Stable expression of luc was observed in skov 3/Luc cells.Ovarian cancer dormancy nude mouse model was successfully established based on the living imaging technology and were verified by the small animals living imaging techniques.Conclusion The ovarian cancer dormancy nude mouse model based on living imaging technology is an ideal experimental animal model to study the mechanism of tumor cell dormancy and the ways to maintain and induce tumor cell dormant.
