丹参水提液对小鼠睾丸生精功能及精子DNA损伤的影响

摘    要:目的 探讨丹参水提液对小鼠睾丸生精功能及精子DNA碎片的影响.方法 将40只昆明小鼠随机均分为低、中、高浓度、空白对照、阳性对照5组,低、中、高浓度组小鼠每天分别灌胃1 500、3 000、6 000 mg/kg丹参水提液,空白对照组每天灌胃10 mL/kg生理盐水,阳性对照组一次性注射100 mg/kg环磷酰胺,30 d后处死小鼠,分别采用流式细胞术、吉姆萨染色法与染色质扩散法检测睾丸生精细胞周期、早期生精细胞微核率及精子DNA碎片率.结果 低、中、高浓度组小鼠睾丸早期生精细胞微核率均低于空白对照组,但差异无统计学意义(P>0.05).随着丹参浓度增加,低、中、高浓度组睾丸单倍体细胞(1C)比例呈上升趋势,且高浓度组显著高于空白对照组(P<0.05);空白对照、低、中、高浓度组睾丸二倍体细胞(2C)、四倍体细胞(4C)、1C∶2C、1C∶4C比例比较均差异无统计学意义(P>0.05).空白对照、低、中、高浓度组小鼠精子DNA碎片率逐渐下降,且高浓度组显著低于空白对照组(P<0.05).结论 丹参水提液对小鼠睾丸生精功能有促进作用,且可减少生精细胞早期染色体畸变与精子DNA损伤.

Abstract:Objective To investigate the effects of Salvia miltiorrhiza water extract on testicular spermatogenic function and sperm DNA damage in mice.Methods Forty Kunming mice were randomly divided into low,medium,and high concentration,control and positive control groups.Mice in low,medium and high concentration groups were given with 1 500 mg/kg,3 000 mg/kg and 6 000 mg/kg salvia miltiorrhiza water extraction by gavage once a day,respectively;as the replaced saline and bolus cyclophosphamide were given to those in control group and positive control group,respec tively.All mice were sacrificed after 30 days,testicular spermatogenic cycle,micronucleus rate and sperm DNA fragmentation were detected by flow cytometry,giemsa staining and chromatin dispersion methods.Results Micronucleus rates of early spermatogenic cells of low,medium and high concentration groups were lower than control group,but with no statistical differences (P > 0.05).With the increase of salvia miltiorrhiza,testicular haploid ratio (1 C) rised gradually,and significant differences were observed between high concentration and control group (P < 0.05).There was no significant difference in testicular diploid (2C),tetraploid (4C),(2C∶ 4C) or (1C∶ 4C) ratio among control,and low,medium,and high concentration groups (P > 0.05).Sperm DNA fragmentation rate decreased gradually among control,and low,medium and high concentration groups,and significant difference was observed between high concentration and control groups (P < 0.05).Conclusion Salvia miltiorrhiza water extract could promote testicular spermatogenic process,reduce spermatogenic chromosome aberration and sperm DNA damage.
