摘    要:目的 探讨miR-135b对子宫内膜癌中HOXA10基因表达的调控作用.方法 RT-PCR方法检测28例子宫内膜癌组织及相应癌旁组织中miR-135b及HOXA10的表达;用miR-135b模拟物及抑制物转染RL-952子宫内膜癌细胞株,RT-PCR检测转染效果并检测FOXO1 mRNA的变化,划痕实验检测转染后对细胞迁移的影响,流式细胞技术检测转染对细胞增殖的影响,Western blot检测HOXA10蛋白的变化.结果 miR-135b在子宫内膜癌组织中较对应的癌旁组织表达升高(P<0.05),HOXA10则降低(P<0.05),经miR-135b模拟物转染后,子宫内膜癌细胞中HOXA10 mRNA和蛋白的表达水平下降(P<0.05);经miR-135b抑制物转染后,子宫内膜癌细胞中HOXA10 mRNA和蛋白的表达水平升高(P<0.05);miR-135b能促进子宫内膜癌细胞的增殖(P<0.05).结论 miR-135b 在子宫内膜癌发生发展中有一定作用,子宫内膜中HOXA10的异常表达受miR-135b的调控.

Abstract:Objective To explore the effects of miR-135b on HOXA10 in endometrial cancer.Methods The expression of miR-135b and HOXA10 in human endometrial cancer tissues and their para-carcinoma tissues was assessed by real-time PCR.miR-135b mimics (miR-135b up-regulated expressions) and miR-135b inhibitors (miR-135b down-regulated expression) were transfected into human endometrial cancer RL-952 cells by liposome method.The transfection efficiency was detected by RT-PCR.Wound healing assay was performed to detect the cell migration ability.Cell proliferation and cell cycle were analyzed 48h after transfection by flow cytometry.The expression of HOXA10 mRNA and protein were assessed by RT-PCR and Western blot.Results The expression of miR-135b in human endometrial cancer tissues was significantly higher than their para-carcinoma tissues (P<0.05).Expression of HOXA10 in endometrial carcinoma tissues was significantly lower than their para-carcinoma tissues (P<0.05).Compared with NC group, the mRNA and the protein expression of HOXA10 was significantly down-regulated in miR-135b mimics group (P<0.05).However, in miR-135b inhibitors group, the mRNA and the protein expression of HOXA10 was significantly up-regulated (P<0.05).MiR-135b can promote the proliferation of endometrial cancer cells (P<0.05).Conclusion MiR-135b plays an important role in the occurrence and development of endometrial carcinoma by regulating HOXA10.
