经脾实质胰岛移植治疗糖尿病小鼠的实验研究

摘    要:目的介绍一种经脾实质内胰岛移植治疗糖尿病小鼠的方法并对移植前后小鼠的血糖、口服糖耐量和组织学进行评价。方法将分离纯化的Balb/c小鼠胰岛培养6 h后,用自制的移植工具进行同种糖尿病小鼠的脾实质内胰岛输注,移植当量为(500±30)IEQ。对移植前后的小鼠血糖进行测定,并在移植后第10天进行糖耐量试验;取受体小鼠脾脏,进行HE染色和免疫组化染色,观察胰岛细胞团在脾内的存活情况。结果受体小鼠移植胰岛后均能维持正常血糖时间达1个月以上,糖耐量试验结果显示与正常小鼠统计学无差异(P=0.69),组织学结果显示胰岛细胞团在小鼠脾实质内存活良好,胰岛有功能,但结构略有松散。结论本研究探索了胰岛在脾实质内移植后的特点,为研究脾实质作为胰岛移植的位点提供了参考。

Abstract:Objective To introduce a method about how to transplant islets into spleen parenchyma in mice, and to evaluate the blood glucose, glucose tolerance and histological stain after transplantation in mice. Methods Balb/c mice were used in this experiment. The (500±30) IEQ purified islets were injected into mice parenchyma by a modified instrument after cultured for 6 h. Then the blood glucose was tested regularly for 1 month, the oral glucose tolerance test was done at the 10th day after islet transplantation, and spleen sample was collected at the 10th day after islet transplantation. The HE and immunohistochemistry staining were carried out to evaluate the is-let morphology and function. Results Mice recipient could achieve and maintain normal blood glucose for more than 1 month after islet transplantation. OGTT results at 10th day after islet transplantation showed that, compared with those in normal mice, the blood glucose levels in mice after islet transplantation were not significant differ-ent (P=0.69). And the immunohistochemical staining showed that the activity and the function of islets was good, while the structure of islet was a bit loose. Conclusion Here we explore the characteristics of pancreatic islets transplantation in spleen parenchyma, which provides a reference for the research on spleen parenchyma as a site for islet transplantation.
