气管内应用GM—CSF重组腺病毒对实验性肺转移癌的治疗作用

摘    要:目的 研究气管内应用GM-CSF重组腺病毒(AdGM-CSF)对实验性肺转移癌的治疗作用。方法 (1)小鼠气管内商入AdGM-CSF,FLISA法测定局部及外周血GM-CSF及相关因子的水平。(2)C57BL/6小鼠尾静脉注射Lewis肺癌3LL细胞建立肺转移癌模型,观察气管内应用AdGMJ-CSF对实验性肺转移癌的治疗作用,包括肺转移结节、动物存活期、生存率变化等,并比较NK细胞和CTL活性的不同。结果 (1)气管内应用AdGM-CSF后72h肺灌洗液中检测到GM-CSF且高于外周血水平,对照组灌洗液及外周血中均未检测到。(2)气管内滴入AdGM-CSF对实验性肺转移癌有治疗作用,可使肺转移结节减少,动物的生存期延长及存活率升高,同时对NK细胞、CTL活性有增强作用。结论 气管内应用GM-CSF重组腺病毒可有效表达并对实验性肺转移癌有治疗作用。

Abstract:Objective To investigate the therapeutic effect of intratracheal injection of recombinant adenovirus GM CSF gene in the treatment of experimental lung metastasis. Methods (1)The concentrations of GM CSF in pulmonary lavage and peripheral blood were determined by ELISA at different time after intratracheal injection of recombinant adenovirus GM CSF. (2)The pulmonary metastatic nodules', survival period, and survival rates were also studied in C57BL/6 mice model with NK activity and CTL activity determined by 51 Cr 4 hour release method. Results (1)After intratracheal recombinant adenovirus GM CSF, the concentrations of GM CSF in the pulmonary lavage was higher than that in the peripheral blood, and GM CSF could not be detected as in the above in the control groups. (2)Intratracheal recombinant adenovirus GM CSF had significant therapeutic effect in the experimental animals with lung metastasis, showing increased CTL and NK activity, longer survival period and higher survival rates compared with those of the control group. Conclusion The study shows that intratracheal adenovirus vector containing GM CSF gene can be effective in experimental pulmonary metastasis through direct intratracheal route.
