高压氧对缺血缺氧性脑损伤幼鼠海马超微结构及主动回避反应的影响

摘    要:目的 观察高压氧治疗对新生大鼠缺血缺氧损伤后脑超微结构和在青春期对主动回避反应的影响.方法 出生7 d的Wistar大鼠制作缺血缺氧性脑损伤模型,采用随机数字表法分为高压氧治疗组和未治疗组,并设正常对照组,采用穿梭箱主动回避行为模型进行主动回避反应(AAR)及被动回避反应(PAR)测试,并观察各组幼鼠海马超微结构.结果 高压氧治疗1次后24 h的幼鼠脑重量较缺血缺氧后不治疗组脑重明显轻.缺氧缺血后海马超微结构变化明显,经高压氧治疗后,海马神经元和突触无明显异常.高压氧治疗组无论是主动回避反应还是被动回避反应均比未治疗组成功率高,两组比较AAR及PAR都有显著性差异(P<0.05,P<0.01).结论 高压氧治疗可减轻缺氧所致脑损伤,有利于受损幼鼠认知能力的恢复.

Abstract:Objective To investigate the effect of hyperbaric oxygen treatment(HBOT) on active avoidance response of neonate rats with hypoxic-ischemic injure.Methods Forty Wistar rats of 7 days old were established into hypoxic-ischemic injury,and were randomly divided into HBOT group and no-HBOT group.Ten normal rats served as normal controls.AAR and PAR of rats were measured in shuttle box.The hippocampal ultrastructure was detected with electron microscope.Results Brain weight of rats underwent HBOT once after 24 h were decreased significantly as compared with no-HBOT group.The hippocampal ultrastructure after hypoxic-ischemic injure changed obviously.Nerve cells were of puff and intumesce.In kytoplasm,rough endoplasmic reticulum and Golgi saccule were expanding and vacuolated,and ridge of bioblast was blur.Regardless of active avoidance response(AAR) or passive avoidance response(PAR),the achievement ratio of HBOT group increased significantly.AAR and PAR between the two groups were of significant difference(P<0.05,P<0.001).Conclusion HBOT may relieve the cerebral anoxic injury and be conducive to cognitive ability of injured immature rats.
