流行性出血热病毒在柏氏禽刺螨体内生存空间和时间的初步研究

摘    要:将感染性柏氏禽刺螨进行冰冻切片,以IFAT检测,结果首次证明革螨的肠组织及生殖器官(卵巢、及子宫孕卵)等部位的细胞内有流行性出血热病毒(EHFV)特异性荧光颗粒,而对照组则阴性。两个感染后的柏氏禽刺螨群体,一个是在第一若螨期感染的,另一个是感染成螨后,挑出其第2代若螨,均在适宜条件下饲养,不仅在饲养15天、25天叮刺乳鼠获阳性结果,而且在158天时将螨洗涤、研磨制成悬液接种乳鼠后,经IFAT检测鼠肺,也均获阳性结果。首次证明此螨群体能维持EHFV158天(约5个月)以上。以上的试验从螨体组织切片上和感染后进行繁殖的螨群体中证明EHFV的存在和维持,都说明此螨确系EHFV的适宜媒介和贮存宿主。

Study on Distribution and Life-Span of Epidemic Haemorrhagic Fever Virus in Ornithonyssus Baooti

Abstract:Ornithonyssus bacoti mites were infected with epidemic haemorrhagic fever virus (EHFV) by biting infected suckling mice. In 26 days afterward, freezing sections of the mites were made and examined by IFAT. For the first time,we detected the specific fluorescent granules of EHFV in both gut cells and tissue cells of the genital organs of O.bacoti, while the controls were negative. We studied two colonies of O.bacoti: one of them was infected with EHFV in protonymph stage, and reared under favorable conditions, and the other, was infected in adult stage, and their 2nd generation protonymph had been reared under the same conditions. We had confirmed that they could transmit EHFV to healthy suckling mice by bitin on the 15th and 25th day.In this article we present the study in which the mites were washed, ground, and emulcified, and then inoculated into healthy suckling mice when the colonies had been reared for 158 days. The results obtained from both colonies were positive, when the lung sections of mice were examined by IFAT. So we have, for the first time, confirmed that EHFV can be maintained in colonies of O. bacoti for at least 158 days (about 5 monthes), either infected at protonymph or adult stages. These results support our previous reports on the evidence of biting, trannsstadial and transovarian passage of EHFV in O. bacoti. We believe that they may be the vector and reservoir of urban rodent type of EHF.
