胰腺癌组织的平均微血管密度和血管内皮细胞生长因子的表达

摘    要:目的:通过检测胰腺癌、慢性胰腺炎及正常胰腺标本中的平均微血管密度(MVD)及血管内皮细胞生长因子(VEGF)表达水平,了解胰腺癌肿瘤血管生成情况.方法:胰腺癌标本26例,慢性胰腺炎标本7例,癌旁正常胰腺组织26例(采用胰腺癌标本中无肿瘤浸润的组织),所有标本均行免疫组化染色.MVD检测为在200倍显微镜下计数5个视野最高数目的微血管,以其平均数表示.VEGF的定量标准为VEGF染色强度与阳性细胞百分比分值的乘积.统计分析采用t检验和二元变量相关分析法.结果:VEGF在胰腺癌、慢性胰腺炎及癌旁正常胰腺组织中均有表达.胰腺癌组织的MVD值及VEGF表达水平均明显高于慢性胰腺炎及癌旁正常胰腺组织(P<0.01).26例胰腺癌组织MVD值及VEGF表达水平在统计学上有相关性(Spearman法r=0.862,P<0.000;Pearson法r=0.855, P<0.000).结论:胰腺癌的微血管生成水平高于慢性胰腺炎及癌旁正常胰腺组织;胰腺癌组织内MVD值及VEGF表达水平在统计学上有相关性.

Abstract:Objective:To detect the microvascular density(MVD) and expression of vascular endothelial growth factor(VEGF) in specimens of pancreatic adenocarcinoma, chronic pancreatitis and normal pancreas so as to understand the angiogenesis of pancreatic adenocarcinoma. Methods: Specimens of pancreatic adenocarcinoma(n=26),chronic pancreatitis(n=7),and paracarcinoma tissue(n=26) were studied immunohistochemically. MVD was determined as the mean of microvessel under 5 microscopic vision fields(200). Quantitative standard of VEGF protein was the product of VEGF staining intensity and positive cellular ratio. Data were analyzed statistically by t-test and Bivariate correlations. Results: Expression of VEGF was detected in pancreatic adenocarcinoma,chronic pancreatitis and normal pancreas. MVD and VEGF expressions of pancreatic adenocarcinoma were statistically higher than those of chronic pancreatitis and normal pancreas(P<0.01). Spearman(r=0.862,P<0.000) and Pearson(r=0.855,P< 0.000) analysis showed that MVD and VEGF expressions in 26 pancreatic adenocarcinoma were statistically correlated. Conclusion: Microvessel angiogenesis in pancreatic adenocarcinoma is statistically higher than that in chronic pancreatitis and normal pancreas. MVD is statistically correlated with VEGF expression in pancreatic carcinoma.
