环氧合酶-2在bFGF诱导的人晶状体上皮细胞增殖中的表达

摘    要:目的检测环氧合酶-2(cyclooxygenase,COX-2)在人晶状体上皮细胞(human lens epithelium cell,HLEC)中的表达,探讨COX-2在后发性白内障(PCO)形成中的意义。方法细胞传代培养,取第三代细胞用于实验。用SP免疫组化法检测bFGF刺激6小时后COX-2蛋白的表达,并与对照组相比较,以胞浆内出现棕黄色颗粒为阳性表达;采用RT—PCR和Western—blot方法检测bFGF刺激下,不同时间点(1h、3h、6h、12h、24h)COX-2的mRNA和蛋白的表达。结果检测的人晶状体上皮细胞中,实验组的细胞COX-2mRNA和蛋白表达均为阳性。且表达呈时间依赖性,随着时间的延长COX-2表达逐渐增加,6h达最高峰;正常对照组表达弱或无表达。结论bFGF可以刺激晶状体上皮细胞增殖,并诱导环氧合酶-2的表达,提示COX-2与人晶状体上皮细胞的增殖密切相关。COX-2可能参与PCO的形成。

Abstract:Objective To measure the expression of cyclooxygenase-2(COX-2)in human lens epithelial cell(HLEC)and to explore the significance of COX-2 in the formation of posterior capsule opacification(PCO).Methods Cell was subculture and the third-generation cell was used for the experiment.The expression of COX-2 protein stimulated by bFGF for 6 hours was measured by SP immunohistochemistry and brown granula were positively expressed in cytoplasm in comparison with the control group.The expression of COX-2 mRNA and protein stimulated by bFGF were measured by RT-PCR and Western-blot at different time periods.Results Cell in test group expressed COX-2 and protein positively,with the characteristic of time dependence.The expression of COX-2 became more significantly as time prolonged,hitting the peak after 6 hours.By comparison,the expression in control group was slight or negative.Conclusions bFGF can induce the expression of COX-2 in human lens epithelial cells;COX-2 has great relation with the proliferation of human lens epithelial cells.Therefore,COX-2 may be involved in the formation of PCO.
