骨髓基质干细胞对急性脊髓损伤大鼠模型神经功能修复的影响

摘    要:目的探索骨髓基质干细胞(bone marrow stromal cells,BMSCs)体外定向诱导分化为神经细胞后移植治疗急性脊髓损伤动物模型的疗效。方法对BMSCs进行体外诱导,并鉴定。将模型大鼠分为3组,将诱导及未诱导的BMSCs和生理盐水分别注入模型鼠脊髓半切点附近,在3个月内用BBB评分法对治疗情况进行评价。结果BMSCs体外诱导后的细胞经免疫组化检测可证实为神经细胞;进行细胞移植的两组动物死亡率明显下降,神经功能有明显恢复(P〈0.05);并且BMSCs诱导后移植组神经功能恢复较未诱导组明显(P〈0.05);而生理盐水组移植前后评分变化不明显。结论BMSCs诱导组治疗脊髓损伤模型的疗效总体好于未诱导的BMSCs组,前两组的疗效均好于生理盐水组。

Abstract:Objective To study curative effects of neural cells transplantation induced in vitro from bone marrow stromal cells(BMSCs) on rat models with injured spinal cord.Methods BMSCs were induced and identified in vitro.The rat models were allocated randomly into 3 groups.Induced BMSCs,non-induced BMSCs and physiological saline were injected into around the site of semisection of spinal cord of the models respectively.Basso,Beattie and Bresnahan(BBB) method was used to assessment of the therapeutic outcome 3 months after transplantation.Results The in vitro post-induction BMSCs were confirmed to be the neural cells by immunocytochemical.An obvious decrease of mortality and a statistically significant improvement in neurological function outcome were noted in rats with BMSCs transplantation groups,and which were more obvious in induced group than non-induced group(P<0.05);however,BBB score of normal saline group had no obvious change after transplantation(P<0.05).Conclusion BMSCs induced in vitro had better effects in treating injured spinal cord model rats compared with that of BMSCs non-induced group,but physiological saline group had worse curative effect than the two BMSCs groups.
