抑制性杀伤细胞受体在NK细胞和T细胞上的表达及其与急性移植物抗宿主病的关系

摘    要: 目的 研究异基因造血干细胞移植(allo-HSCT)患者移植前后外周血NK及T细胞上4种抑制性杀伤细胞受体(CD158a、CD158b、NKB1和CD94/NKG2A)的表达及其与急性移植物抗宿主病(aGVHD)的关系。方法 采用流式细胞术检测NK及T细胞上抑制性杀伤细胞受体的表达。结果 NK细胞上CD158a和CD158b的表达于移植后3~4个月、NKB1和CD94/NKG2A的表达于移植后2个月恢复移植前水平。移植前后CD158a和NKB1在CD+3 T细胞上持续低水平表达;移植前CD158b和CD94/NKG2A在CD+3 T细胞上的表达水平较高,且主要表达于CD+8 T细胞上,移植后其在CD+8 T细胞上的表达增加。CD+8 T细胞上CD158b的表达在发生Ⅰ度aGVHD时显著增高,与无aGVHD组及Ⅱ~Ⅳ度aGVHD组相比,差异均有统计学意义(P均<0.05);在发生Ⅱ~Ⅳ度aGVHD时增高不明显,与无aGVHD组相比,差异无统计学意义(P>0.05)。结论 CD158b在CD+8 T细胞上高表达可能有助于降低T细胞同种反应性,减轻aGVHD的程度。

Abstract:Objective To investigate the expression of four type natural killer(NK) cells inhibitory re-ceptors (CD158a, CD158b, NKB1 and CD94/NKG2A) on peripheral blood NK cells and T lymphocytes in patients before and after allogeneic hematopoietic stem cell transplantation (allo-HSCT) and the relationship with acute graft-versus-host disease(aGVHD). Methods Flow cytometry analyzes the expression of four type NK cells inhibitory receptors on NK cells and T cells. Results The expression of CD158a and CD158b on NK cells was recovered 3~4 months after allo-HSCT, while NKB1 and CD94/NKG2A was recovered 2 months after allo-HSCT. The percentage of CD158a and NKB1 expressing on CD3 positive T cells was low and there was little change after allo-HSCT. The percentage of CD158b and CD94/NKG2A expressing on CD3 positive T cells be-fore transplantation was relatively higher compared with other NK cells inhibitory receptors, such as CD158a and NKB1, and mainly on CD8 positive T cells. The expression of CD158b and NKG2A on CD8 positive T cells was increased after allo-HSCT. The percentage of CD158b expressing on CD8 positive T cells was significantly in-creased in patients with grade I aGVHD compared with those without aGVHD and those with grade Ⅱ~ Ⅳ aGVHD (P < 0.05, respectively), while no significantly increased in patients with grade Ⅱ~ Ⅳ aGVHD com-pared with no aGVHD (P > 0.05 ). Conclusion The high expression of CD158b on CD8 positive T cells may fa-vor to control T cells alloreactivity and reduce aGVHD.
