原发性胆囊癌癌组织中DCs的浸润和IL-10表达的研究

目的研究原发性胆囊癌(PBC)组织中树突状细胞(DCs)的浸润和白细胞介素-10(IL-10)的表达情况,探讨PBC组织局部免疫状态变化与其不良预后之间的关系。方法回顾性分析我院56例胆囊手术标本,采用连续切片法及免疫组织化学(IHC)染色,检测树突状细胞(DCs)(联合单抗CD1a和S-100标记)的浸润和IL-10的表达。结果以CD1a和S-100为标志的DCs在PBC癌组织中的浸润比其在慢性胆囊炎、胆囊腺瘤组织中的浸润下降(P<0.05),并与其分化程度、浸润及淋巴转移有关(P<0.05);IL-10的表达与病变性质有关,但与PBC的进展无关。结论PBC的发生发展可能与病人自身以DC为代表的免疫功能下降与抑制有关。IL-10可能有抑制DC功能的作用。

Infiltration of DCs and expression of IL-10 in primary gallbladder cancer

Objective To study the infiltration of dendritic cells(DCs) and expression of interleukin-10(IL-10) and their roles in antitumor immunity in primary gallbladder cancer.Methods There were 56 specimens of various lesions of gallbladder.Their monoclonal antibodies against cell markers,S-100,CD1a,and IL-10 were determined by immunohistochemistry.The infiltration of DCs and expression of IL-10 in various lesions of gallbladder epithelia were quantitively analyzed.Results The percentages of S-100 and CD1a positive DCs in gallbladder cancer were lower than those in other benign lesions of gallbladder(P<0.05),and the number varied with tumor differentiation type,invasion depth and lymph node metastasis(P<0.05);Expression of IL-10 in tumor tissue was higher than that in benign diseases(P<0.05),but not varied with tumor differentiation,invasion depth or lymph node metastasis.Conclusion DCs plays a positive important role,and their function and number in primary gallbladder cancer are a key in initiating tumor specific immunity and may be used to estimate its prognosis.While IL-10 plays a negative role in PBC tumor immunity.
