摘    要:目的:探讨长链非编码RNA(lncRNA)在药物性肝损伤小鼠肝组织中表达谱的变化。方法:利用lncRNA芯片技术检测小鼠正常肝组织和对乙酰氨基酚诱导的药物性肝损伤肝组织中的lncRNA表达谱的变化,通过对原始数据进行预处理达到均一化后,筛选出差异表达的lncRNA并进行分析。结果:与正常肝脏组织比较,其中1.5倍以上变化并且差异有统计学意义(P〈0.05)的lncRNA认为是差异表达的lncRNA。1.5倍以上变化的共68条,1.5倍以上升高的共21条,1.5倍以上降低的共47条;2倍以上升高的共7条,2倍以上降低的共14条;3倍以上升高共2条,3倍以上降低的共5条。结论:药物性肝损伤小鼠肝脏组织与正常小鼠肝脏组织比较,lncRNA表达谱发生了显著变化,提示这些差异性lncRNAs可能参与了药物性肝损伤发生及发展过程。

Study on expression profile of long non-coding RNA in hepatic tissue of druginduced liver injury

Abstract:Objective: To analyze the expression profile variation of long non-coding RNA (lncRNA) in the hepatic tissue of drug-induced liver injury. Methods: lncRNA mieroarray technology was used to inspect the difference of IncRNA expression profile between mice's normal liver and liver injury in mice induced by aeetaminophen, the lncRNA with differential expression after pretreatment and homogenization of raw data were screened out and amalysised. Results: Compare with normal hepatic tissue, and 68 lncRNA with more than 1.5 times variation and significant difference (P 〈 0. 05 ) by statistical analysis were regarded as lneRNA with differential expression, while 21 was increase more than 1.5 times and 47 was reduce more than 1.5 times; 7 was increase more than 2 times and, 14 was reduce more than 2 times; 2 was increase more than 3 times and 5 was reduce more than 3 times. Conclusion: LncRNA expression profile of drug-induced liver injury changes significantly in comparison with normal hepatic tissue. The different lncRNAs may play important roles in the development and progression.
