人口腔黏膜上皮细胞的培养及其在PLGA膜上生长的初步实验

目的 :探讨人口腔黏膜上皮细胞的原代培养和体外生长方法。方法 :采用美国DispaseⅡ试剂获取人体正常口腔黏膜上皮的原代细胞 ,将其种植在PLGA膜上 ,通过光学显微镜形态学检查、扫描电镜和免疫组织化学进行细胞定性。结果 :采用DispaseⅡ可成功地分离出口腔黏膜上皮层和上皮下层 ,通过进一步处理后可获取口腔黏膜上皮细胞的原代细胞 ,将其植入PLAG膜上 ,细胞获得很快生长 ,5d时细胞出现分化 ,8d时可以长到 5层左右。结论 :本方法可较好地获取口腔黏膜原代细胞 ,获取的口腔黏膜原代上皮细胞种植在PLAG膜上可获得良好的生长。

Primary experimental study on the culture of human epithelial cells of oral mucosa and their growth on PLGA membrane

Objective To investigate the method of culturing hu man epithelial cells of oral mucosa and to observe their growth on PLGA membrane . Methods Human epithelial cells were cultured by explants tech nique in vitro, and then were observed under light microscope, and scanni ng e lectron microscope (SEM), and were identified by AE1/AE3 staining; cultured epi thelial cells were seeded on the PLGA membrane for some days. Results Epithelial cells grew well in round, polygon and triangle in culturing m ed ium, the results of light microscope and SEM observation showed that the struct ure of cultured cells were similar to those in normal human oral mucosa. Immunoh istochemical staining showed that the cultured cells were AE1/AE3 positive, the y grew well on PLGA membrane. Conclusion Epithelial cells of hu man oral mucosa were cultured successfully. PLGA membrane can be urged as a scaf fold for culture of epithelial cells of human oral mucosa.
