多指标综合评分法优选华蟾素提取液醇沉工艺的研究

摘    要:目的研究并确定华蟾素提取液的最佳醇沉工艺参数,为华蟾素醇沉工艺的产业化提供参考。方法采用单因素试验法分别考察初膏相对密度、终点乙醇含量、乙醇加入速度、初始乙醇浓度、静置温度和药液pH等因素对华蟾素保留率的影响。在单因素实验结果的基础上,设计L9(34)正交实验,以华蟾素保留率、杂质去除率和华蟾素纯度为指标,采用多指标综合评分法确定华蟾素醇沉的最佳工艺参数。结果华蟾素提取液醇沉的最佳工艺条件为:终点乙醇含量75%,初膏相对密度1.10,所用乙醇浓度82%,静置温度10℃。结论该方法稳定、可行,具有很大的工业化应用价值。

Abstract:ObjectiveTo investigate the influences of different parameters on the process of alcohol precipitation and to assure the optimal conditions in this study. MethodsThe influences of the concentration of alcohol,the relative density of herb liquor,the speed of adding alcohol and setting temperature on the retenthor rate of active components in Cinobufacini alcohol precipitation process were investigated by single factor experiment.Then an orthogonal test was conducted by the four factors with greater influence.The optimization of technology was performed by using orthogonal design and multi-index compositive score method. ResultsThe optimal process parameters were the relative density of herb liquor 1.10,the final concentration of alcohol 82%,the concentration of alcohol 75% and the settling temperature 10℃. ConclusionThe method is stable and feasible,and has widely industrial application prospect.
